摘 要:近年来,纳米载体因其优异的负载能力、高效的递送性能以及精准的靶向性,在药物及功能性核酸的递送领域受到了广泛关注.这些特性使纳米载体能够将药物或核酸直接输送至病变区域,为癌症治疗提供了新的策略.为了降低纳米药物对正常组织的毒副作用,研究人员致力于开发刺激响应型纳米载体,进而实现时空可控的药物或诊疗平台的激活,并进行靶向释放.能够激活响应型纳米载体的刺激主要分为外源性物理刺激和内源性生理刺激,均能够使纳米载体发生结构和性质的改变,从而实现精准的药物递送,并进行相关癌症治疗.文章主要对响应型纳米载体的分类、构建原理和在癌症治疗中的应用进展进行了介绍,并对响应型纳米载体的未来应用进行了展望.Recently,nanocarriers have received widespread attention in the field of drug and functional nucleic acid delivery due to their excellent loading capacity,efficient delivery performance,and precise targeting.These characteristics enable nanocarriers to directly deliver drugs or nucleic acids to the affected area,providing new strategies for cancer treatment.for facilitating the treatment of diseases like cancer.To mitigate the toxic side effects of nanomedicines on normal tissues,researchers have dedicated to developing stimuli-responsive nanocarriers,aiming to achieve spatiotemporal controllable activation of drugs or theranostic platforms and targeted release.The stimuli that can activate responsive nanocarriers are mainly divided into exogenous and endogenous physiological ones,both of which can induce structural and characteristic changes in nanocarriers,thereby enabling precise drug delivery and enhanced treatment.This review mainly introduces the classification,construction principles,and application progress of responsive nanocarriers in cancer treatment,and provides a perspective on their further applications.
