电针通过调节MIP-1α/CCR1/CCR5信号通路对大鼠带状疱疹后神经痛的机制研究

摘  要:目的 基于趋化因子配体3(CCL3,又称MIP-1α)/C-C趋化因子受体(CCR)1/CCR5信号通路探讨电针治疗大鼠带状疱疹后神经痛的机制。方法 将80只SPF级大鼠(雌雄不限)随机分为空白组、模型组、普瑞巴林组、电针组,每组20只。除空白组外,其余各组大鼠经腹腔注射200μg/kg树胶脂毒素混合液建立带状疱疹后神经痛模型。造模成功后,空白组大鼠予0.9%氯化钠灌胃;普瑞巴林组予27 mg/(kg·d)普瑞巴林灌胃;电针组取“夹脊”“肝俞”“胆俞”“支沟”“阳陵泉”“合谷”,行电针治疗;每组均干预7 d。比较各组大鼠机械和热痛觉敏感性,HE染色观察脊髓神经元形态变化,ELISA法测定脊髓组织炎症因子白介素(IL)-4、IL-6、肿瘤坏死因子(TNF)-α与神经递质神经激肽(NK)-1、P物质(SP)水平,Western blot检测MIP-1α/CCR1/CCR5信号通路相关蛋白表达。结果 与空白组比较,模型组大鼠机械痛觉敏感性降低、热痛觉敏感性升高(P<0.01),IL-4、IL-6、TNF-α、NK-1、SP、MIP-1α、CCR1、CCR5水平均升高(P<0.01);与模型组比较,电针组与普瑞巴林组大鼠机械痛觉敏感性升高(P<0.01),热痛觉敏感性与上述其他各项指标表达均有明显下降(P<0.01)。结论 电针能够通过调节MIP-1α/CCR1/CCR5信号通路发挥治疗大鼠带状疱疹后神经痛的作用。Objective To explore the mechanism of electroacupuncture in regulating CC-chemokine ligand(CCL3,or MIP-1α)/C-C chemokine receptor(CCR)1/CCR 5 signaling pathway on postherpetic neuralgia in rats.Methods Eighty SPF rats(male and female)were randomly divided into blank group,model group,pregabalin group and electroacupuncture group,with 20 rats in each group.Except for the blank group,rats in the model group,pregabalin group and electroacupuncture group received 200μg/kg of gum lipotoxin mixture intraperitoneally to establish a model of postherpetic neuralgia.After successful modeling,the blank group was given 0.9%sodium chloride by gavage.The pregabalin group were given 27 mg/(kg•d)pregabalin orally.In the electroacupuncture group,"Jiaji""Ganshu""Danshu""Zhigou""Yanglingquan"and"Hegu"were selected for electroacupuncture treatment.And each group was treated for 7 days.The mechanical and thermal pain sensitivity of rats in all groups were compared,the morphological changes of spinal cord neurons were observed by HE staining,and the levels of inflammatory factors IL-4,IL-6,TNF-αand neurotransmitters NK-1 and substance P(SP)in spinal cord tissue were determined by ELISA.The protein expression related to MIP-1α/CCR1/CCR5 signaling pathway was measured by Western blot.Results Compared with blank group,the mechanical pain sensitivity was decreased(P<0.01),while the levels of thermal pain sensitivity,IL-4,IL-6,TNF-α,NK-1,SP,MIP-1α,CCR1 and CCR5 were increased in model group(P<0.01).Compared with model group,the mechanical pain sensitivity increased(P<0.01),while thermal pain sensitivity,as well as the expressions of other indicators above were decreased(P<0.01)in electroacupuncture group and pregabalin group.Conclusion Electroacupuncture can treat postherpetic neuralgia in rats by modulating MIP-1α/CCR1/CCR5 signaling pathway.
