摘  要:本研究旨在建立榴姜(石榴皮-干姜)超微粉的质量控制标准。将石榴皮与干姜按照1∶1的比例混合并制成超微粉,以激光粒度分析法测定榴姜超微粉粒径;采用显微鉴别法观察榴姜超微粉的显微特征;以薄层色谱分析法对干姜、石榴皮进行定性鉴别;采用高效液相色谱法对鞣花酸、6-姜辣素进行含量测定。结果表明,榴姜超微粉粒径均小于48µm;显微镜下未见明显纤维结构;薄层色谱中,供试品与对照药材在相应的位置上均显示相同颜色的斑点,阴性样品无干扰;高效液相色谱中,鞣花酸和6-姜辣素的含量测定方法阴性样品无干扰,两者线性关系系数R^(2)≥0.999,精密度、重复性、系统耐受性、稳定性及含量测定RSD均小于2%,鞣花酸和6-姜辣素平均回收率均高于95%。对10批样品检测后,规定每1g榴姜超微粉中鞣花酸、6-姜辣素含量分别不得低于1.44mg、0.62mg。本试验建立的方法专属性强,重复性良好,易于操作,可用于榴姜超微粉的质量控制。The objective was to establish the quality control standard for the ultrafine powder of Liujiang.Pomegranate peel and dried ginger were mixed in a ratio of 1∶1 to prepare the ultrafine powder.The particle size of Liujiang ultrafine powder was determined using laser particle size analysis;microscopic identification was employed to observe the microscopic characteristics of the powder;thin-layer chromatography(TLC)was used for qualitative identification of dried ginger and pomegranate peel;high-performance liquid chromatography(HPLC)was utilized for the determination of the content of ellagic acid and 6-gingerol.The results showed that the particle size of Liujiang ultrafine powder was less than 48µm after processing;no significant fibrous structure was observed under the microscope;in TLC,the test samples and reference materials showed spots of the same color at corresponding positions,with no interference from negative samples;in HPLC,the content determination methods for ellagic acid and 6-gingerol showed no interference from negative samples,with a linearity correlation coefficient R^(2) of at least 0.999,RSD of precision,repeatability,system suitability,and stability and content determination were all less than 2%,and the average recovery rate of ellagic acid and 6-gingerol were all above 95%.After testing 10 batches of samples,it was stipulated that the content of ellagic acid and 6-gingerol in 1 g of Liujiang ultrafine powder should not be less than 1.44 mg and 0.62 mg,respectively.The established method is specific,reproducible,easy to operate,and can be used for the quality control of Liujiang ultrafine powder.
