基于网络药理学探究增液颗粒治疗便秘的作用机制

摘  要:母猪便秘是母猪妊娠中后期的常见疾病,若治疗不及时,该病可对母猪及仔猪均造成危害。增液颗粒来源于增液汤,经临床验证,该制剂治疗母猪低热、尿少色黄、排便困难等便秘相关症状疗效确切。本文基于网络药理学来探究增液颗粒治疗便秘的药效作用机制。通过TCMSP等数据库搜集增液颗粒的活性成分和关键靶点,筛选与便秘有关的疾病共有靶点,构建PPI网络;使用DAVID数据库,对靶点进行GO功能注释和KEGG富集数据分析;将预测的有效成分与核心靶点进行分子对接验证。结果显示,增液颗粒主要活性成分为甲基麦冬二氢高异黄酮A/B、麦冬二氢高异黄酮A、月桂酸等,与便秘有关的核心靶点主要有酪氨酸蛋白激酶(SRC)、丝氨酸/苏氨酸激酶1(AKT1)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF)、雌激素受体1(ESR1)等。GO功能注释显示,治疗便秘时增液颗粒富集的主要生物学过程有磷酸化、蛋白磷酸化等,细胞组分富集显示大部分靶点与树突、神经元细胞体、神经元投影等相关,分子功能主要富集在蛋白丝氨酸/苏氨酸/酪氨酸激酶活性、蛋白激酶活性、蛋白质结合等。KEGG富集表明,增液颗粒治疗便秘的机制可能与PI3K/AKT、ErbB、癌症等信号通路有关。分子对接结果显示,药物活性成分与核心靶点结合性良好。结果表明,增液颗粒中有效成分甲基麦冬二氢高异黄酮A/B、月桂酸等可通过作用于SRC、TNF、AKT、IL-6等靶点及调控PI3K/AKT等信号通路来改善便秘。Sow constipation is a common disease in the middle and late stages of pregnancy,and if not treated promptly,it can cause harm to both sows and piglets.Zengye granules,derived from Zengyetang,have been clinically proven effective in treating sow-related constipation symptoms such as low fever,scanty and dark urine,and defecation difficulties.Based on network pharmacology,this study explored the mechanism of action for Zengye granules in treating constipation.Active components and key targets were collected through databases such as TCMSP,selecting common targets related to constipation to construct a PPI network.The DAVID database was used to perform GO functional annotation and KEGG pathway enrichment analysis on the targets.Molecular docking verification was conducted between the predicted active ingredients and core targets.The results showed that the main active ingredients of Zengye granules were methyl ophiopogonanone A/B,ophiopogonanone A,lauric acid,etc.The core targets related to constipation mainly included SRC,AKT1,IL-6,TNF,ESR1,etc.The GO functional results showed that the main biological processes for the enrichment of fluid enhancing particles in constipation included phosphorylation,protein phosphorylation,etc.The cellular composition results showed that most of the targets were related to dendrites,neuronal cell body,neuronal projections,etc.The molecular functions were mainly enriched in protein serine/threonine/tyrosine kinase activity,protein kinase activity,protein binding,etc.The mechanism of treating constipation might be related to pathways in cancer,ErbB and PI3K/AKT signaling pathway.The molecular docking results indicated that the active ingredients of the drug had good binding affinity with the core target of the disease.The effective active ingredients in Zengye granules,such as methyl ophiopogonanone A/B,ophiopogonanone A,lauric acid,etc,could improve constipation by acting on targets like TNF,AKT,IL-6,and regulating signaling pathways such as PI3K/AKT.
