摘 要:采用高效液相色谱法(HPLC)测定中兽药马针颗粒中盐酸小檗碱、盐酸药根碱2种活性成分含量,并进行特征图谱研究,为市售中兽药马针颗粒的质量评估提供参考依据。色谱条件:Eclipse Plus C_(18)色谱柱(4.6 mm×250.0 mm,5µm),检测波长215 nm,以乙腈-0.02 mol/L磷酸二氢钾为流动相,等度洗脱15 min,流速0.7 mL/min,柱温30℃,进样体积10µL。结果显示,在方法学考察中,盐酸小檗碱、盐酸药根碱的线性关系良好,相关系数R^(2)分别为0.9997、0.9996,平均回收率分别为103.18%、100.00%,RSD分别为0.56%、1.10%;建立的马针颗粒特征图谱共确定了3个特征峰,相似度为0.818~1.000。本试验建立的检测方法快速准确,结果可靠,适用于马针颗粒中盐酸小檗碱、盐酸药根碱的含量测定,建立的特征图谱适用于马针颗粒的质量评估。The research aims to establish a method for detecting the content of berberine hydrochloride and jatrorrhizine hydrochloride in Mazhen granules using high-performance liquid chromatography(HPLC)and conduct fingerprint analysis,so as to provide a reference for the quality evaluation of commercially available Chinese veterinary medicine Mazhen granules.High performance liquid chromatography was used,with Eclipse Plus C_(18) column(4.6 mm×250.0 mm,5µm),detection wavelength was 215 nm,acetonitrile-0.02 mol/L KH_(2)PO_(4) was used as the mobile phase with isocratic elution of 15 min at a flow rate of 0.7 mL/min,column temperature was 30℃,and injection volume was 10µL.The results showed that the linear relationship was good in the methodological investigation of berberine hydrochloride(R^(2)=0.9997)and jatrorrhizine hydrochloride(R^(2)=0.9996).Their average recovery was 103.18%and 100.00%with RSD of 0.56%and 1.10%,respectively.A total of three characteristic peaks were identified in the established characteristic chromatogram of Mazhen granules,and the similarity was 0.818-1.000.The detection method established in this experiment is fast,accurate and reliable.It is suitable for the determination of berberine hydrochloride and jatrorrhizine hydrochloride in Mazhen granules.The characteristic fingerprint is suitable for the quality evaluation of Mazhen granules.
