摘 要:目的探讨芳香烃受体(AHR)在EB病毒(EBV)阳性鼻咽癌(NPC)细胞系中的作用。方法应用小干扰siAHR敲低EBV阳性NPC细胞系C666-1细胞中AHR的表达,采用Western blot技术检测C666-1细胞AHR、上皮间质转化(EMT)以及自噬相关蛋白的表达,RT-qPCR技术检测细胞中AHR的mRNA表达,CCK8和Transwell分析检测细胞增殖和迁移能力。结果与转染阴性对照siRNA细胞(对照组)相比,C666-1细胞转染siAHR组AHR蛋白和mRNA表达均下调,差异有统计学意义(t=4.154、9.490,P<0.05);敲低AHR表达能够抑制细胞增殖能力(F=14.49、80.99,P<0.05),减少细胞迁移数量(t=2.855,P<0.05),降低EMT相关蛋白ZEB1、N-Cadherin表达水平并升高E-Cadherin的表达水平(t=3.504~4.073,P<0.05),上调自噬相关蛋白ATG3、ATG5、ATG7和ATG12的表达(t=2.822~6.743,P<0.05)。结论在EBV阳性NPC细胞中,AHR能通过影响EMT相关蛋白表达促进细胞迁移、增强细胞增殖能力,影响细胞自噬相关蛋白的表达,在NPC中发挥促癌作用。Objective To investigate the role of aryl hydrocarbon receptor(AHR)in Epstein-Barr virus(EBV)-positive nasopharyngeal carcinoma(NPC)cell lines.Methods Small-interfering RNAs targeting AHR(siAHR)were used to knock down the expression of AHR in EBV-positive NPC C666-1 cells.Western blot was used to measure the expression of AHR,epithelial-mesenchymal transition(EMT)-related proteins,and autophagy-related proteins in C666-1 cells.The mRNA expression level of AHR was determined by RT-qPCR.The abilities of proliferation and migration of the cells were determined using Cell Counting Kit-8 and Transwell assay.Results Compared with the control group,C666-1 cells transfected with siAHR showed significantly downregulated expression of AHR protein and mRNA(t=4.154,9.490;P<0.05),a significantly inhibited proliferation ability(F=14.49,80.99;P<0.05),a significantly reduced number of migratory cells(t=2.855,P<0.05),significantly decreased expression of EMT-related zinc finger E-box binding homeobox 1 and N-cadherin and increased E-cadherin expression(t=3.504-4.073,P<0.05),and significantly upregulated expression of the autophagy-related proteins ATG3,ATG5,ATG7,and ATG12(t=2.822-6.743,P<0.05).Conclusion In EBV-positive NPC cells,AHR can affect the expression of EMT-related proteins to promote cell migration and proliferation and regulate the expression of autophagy-related proteins,thereby playing a cancer-promoting role.
