摘 要:为研究植物开花整合基因FT(Flowering Locus T)及其同源基因在忍冬(Lonicera japonica Thunb.)开花过程中的表达模式及功能,本研究通过生物信息学方法,利用忍冬品种“华金6号”基因组信息,筛选获得7个LjFT及其同源基因,并对其进行了系统分析。利用实时荧光定量PCR(real-time quantitative PCR,RT-qPCR)分析LjFT及其同源基因的表达情况,发现LjFT基因的表达量在扦插苗和实生苗中随日照时间的增加逐渐上升,表明其可能是忍冬开花的促进子;LjFTL2、LjFTL3、LjFTL4、LjFTL5和LjFTL6基因的表达量在扦插苗和实生苗中随日照时间的增加逐渐下降,表明其可能是忍冬开花的抑制子。进一步利用异源表达拟南芥的方法对LjFT进行功能分析发现,与对照相比,LjFT过表达的拟南芥植株开花时间提前,说明LjFT具有促进植物提早开花的功能。本研究结果为解析忍冬开花时间的调控机制提供了重要线索,同时为不同花期忍冬新品种的选育提供了基因靶点。To investigate the role of the Flowering Locus T(FT)gene and its homologous genes in the process of flowering in Lonicera japonica,this study obtained seven LjFT and its homologous genes from the genome of L.japonica“Huajin 6”and systematically analyzed them using bioinformatics methods.By real-time fluorescence quantitative PCR(RT-qPCR),it was found that the expression of LjFT increased gradually with the prolongation of daylight duration in both cutting seedlings and seed seedlings,indicating that LjFT might act as a promoter of flowering.The expression of LjFTL2,LjFTL3,LjFTL4,LjFTL5 and LjFTL6 gradually decreased with the prolongation of daylight duration in both cutting seedlings and seed seedlings,indicating that they might act as repressors of flowering.Further functional studies on LjFT revealed that compared to the control,over-expression of LjFT in Arabidopsis plants resulted in an earlier flowering time,which suggested that LjFT possessed the capability to promote early flowering in the plants.The findings presented above provided a research reference for further research on the molecular mechanisms of flowering and breeding new varieties of L.japonica with different flowering time.
