摘  要:目的探讨不同剂量四君子汤颗粒对多囊卵巢综合征大鼠(PCOS)炎症状态和脂代谢的影响。方法选取30只3周龄雌性SD大鼠,随机分为对照组(CON组,n=6)和PCOS造模组(n=24),造模组采用来曲唑(1mg/kg·d,溶于0.5%羧甲基纤维素钠溶液)灌胃联合高脂饲料喂养,CON组采用等量0.5%羧甲基纤维素钠溶液灌胃,普通饲料喂养,连续28d。造模成功后将造模组分为PCOS组、四君子汤低剂量组(L-SJZD组)、中剂量组(M-SJZD组)和高剂量组(H-SJZD组),每组6只,四君子汤组分别灌胃5.25g/kg·d、10.5g/kgd、21g/kg·d四君子汤颗粒,PCOS组与CON组灌胃等量生理盐水,均以普通饲料喂养,连续21d。记录大鼠动情周期,称重大鼠体质量及卵巢并计算卵巢指数,HE染色法观察卵巢组织形态学变化,酶联免疫吸附试验(ELISA)测定大鼠血清睾酮(T)、黄体生成素(LH)、卵泡刺激素(FSH)水平,Westernblot法检测卵巢组织中白介素-1β(IL-1β)、白介素-18(IL-18)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)、白介素-4(IL-4)蛋白,全自动生化分析仪测定大鼠血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)水平。结果与CON组比较,PCOS造模组大鼠动情周期紊乱,卵巢呈多囊样改变,卵巢指数升高(F=20.700,P<0.05),血清T、LH升高(t=22.740,t=43.040,P<0.05),FSH降低(t=9.264,P<0.05),提示造模成功。经四君子汤颗粒治疗后,大鼠动情周期恢复,卵巢多囊样变化改善;与PCOS组比较,各治疗组大鼠血清T、LH、LH/FSH均降低(F=231.100,F=483.100F=200.900,P<0.05),FSH均升高(F=66.930,P<0.05),M-SJZD组及H-SJZD组T、LH、LH/FSH均低于L-SJZD组(P<0.05),FSH均高于L-SJZD组(P<0.05)。与CON组比较,PCOS组大鼠卵巢组织促炎因子IL-1β、IL-18、IL-6、TNF-α和CRP蛋白表达水平上调(F=116.000,F=46.960,F=166.200,F=185.500,F=66.150,P<0.05),而抗炎因子IL-4蛋白表达下调(F=154.100,P<0.05);与PCOS组比较,各治疗组大鼠卵巢组织Objective To investigate the ffects of diferent doses of Si Jun Zi Decoction granules on the inflammatory status and lipid metabolism in rats with polycystic ovary syndrome(PCOS).Methods Thirty 3-week-old female SD rats were randomly divided into a control group(CON group,n=6)and a PCOS modeling group(n=24).The PCOS modeling group received gavage of letrozole(1 mg/kg d,dissolved in 0.5%carboxymethyl cellulose sodium solution)combined with a highfat diet for 28 consecutive days,while the CON group was administered an equivalent volume of 0.5%carboxymethyl cellulose sodium solution and regular diet.Following successful modeling,the PCOS group was further divided into a PCOS group,a low-dose Si Jun Zi Decoction group(L-SJZD group),a medium-dose group(M-SJZD group),and a high-dose group(H-SJZD group),with 6 rats in each group.The Si Jun Zi Decoction groups received gavage of 5.25 g/kg d,10.5 g/kg d,and 21 g/kg·d of Si Jun Zi Decoction granules,while the PCOS and CON groups were administered an equivalent volume of saline,all maintained on a regular diet for 21 days.Estrous cycles were recorded,ovarian morphological changes were observed through HE staining,the ovarian index was measured and recorded,and serum levels of testosterone(T),luteinizing hormone(LH),and follicle-stimulating hormone(FSH)were measured using ELISA.Western blot analysis was employed to detect the protein expressions of interleukin-1β(IL-1β),interleukin-18(IL-18),interleukin-6(IL-6),tumor necrosis factor-alpha(TNF-α),C-reactive protein(CRP),and interleukin-4(IL-4)in ovarian tissues.A fully automatic biochemical analyzer was used to measure serum cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)levels.Results The PCOS modeling group exhibited disrupted estrous cycles,and HE staining revealed polycystic changes in the ovaries.Compared to the CON group,the ovarian index of PCOS group was increased(F=20.700,P<0.05),serum levels of T and LH were elevated(t=22.740,t=43.040,P
