摘  要:目的通过豚鼠局部封闭涂皮实验和小鼠局部淋巴结法研究3,4-二硝基呋咱基氧化呋咱[3,4-bis(3-nitrofurazan-4-yl)furoxan,DNTF]的皮肤致敏性。方法以2,4-二硝基氯苯为阳性对照,采用豚鼠局部封闭涂皮实验(BT法)和局部淋巴结法(local lymph node assay,LLNA),对DNTF进行致敏性研究。结果豚鼠染毒24和48 h后致敏率分别为35%和50%,中度致敏性;LLNA:DA法检测显示刺激指数(stimulus indexes,SI)>1.8,中度致敏物;LLNA:BrdU-ELISA法检测显示SI>1.6,中度致敏物。结论DNTF具有致敏性,为中等致敏物。为保护接触者的身体健康提供基础数据,同时为小鼠局部淋巴结法在国内化学品毒性评价中的发展和应用积累了实验数据。Objective To detect skin sensitization of 3,4-Bis(3-nitrofurazan-4-yl)furoxan(DNTF)by a local lymph node assay in mice and the occluded patch test of Buchler(BT).Methods According to BT test and local lymph node assay(LLNA)standard method,2,4-dinitrochlorobenzene was used as the positive control to detect sensitization of DNTF.Results The result from the BT test show that DNTF was a moderate sensitizer.The result from LLNA test show that the stimulus indexes of 10%DNTF were SI>1.8,and SI>1.6,and DNTF was a moderate sensitizer.Conclusion DNTF can cause skin sensitization,it is a moderate sensitizer.This study might provide basic data for protecting the health of contacts,and accumulate experimental data for the development and application of local lymph node method in chemical toxicity evaluation in China.
