摘  要:目的研究一株Wa-like中国G3P[8]-I5-A8基因型RVA重配株全基因组特征,并分析与RVA疫苗株的抗原表位之间差异。方法采用反转录-聚合酶链式反应(RT-PCR)方法,对2023年中国青岛采集的一份G3P[8]基因型RVA标本进行全序列扩增。利用在线RVA自动分型工具(http://rotac.regatools.be)对测序结果进行分型,应用MEGA 11.0、Geneious Prime 2023.2.1、Geneious 9.0.2和DNASTAR 5.1等软件对RVA毒株进行全序列遗传变异和进化分析。结果成功获取一株G3P[8]基因型RVA(QD01)的全基因组序列,该毒株具有独特的类似Wa-like的基因型:G3-P[8]-I5-R1-C1-M1-A8-N1-T1-E1-H1。与RVA疫苗株RotaTeq^(TM)(G3)、LLR和Rotarix的VP7中和抗原表位对比有4~13个差异且均在抗原表位的关键位置7-1 b差异较大。结论本研究RVA毒株QD01为G3-P[8]-I5-A8基因型,遗传进化分析推测该毒株可能是亚洲地区猪源RVA和人源RVA毒株间发生重配产生的新型毒株,QD01与RVA疫苗株在抗原表位的关键位置差异较大。Objective To study the genome-wide characteristics of a Wa-like Chinese G3P[8]-I5-A8 genotype rotavirus(RVA)reassortant strain and analyze the differences in antigenic epitopes between the and RVA vaccine strain.MethodsReverse transcription-polymerase chain reaction(RT-PCR)was used to amplify the complete genome sequence of a RVA G3P[8]genotype strain from the specimen collected in Qingdao,China,in 2023.The sequencing result were typed using the online RVA automatic typing tool(http://rotac.regatools.be).The software,including MEGA 11.0,Geneious Prime 2023.2.1,Geneious 9.0.2 and DNASTAR 5.1 were applied to analyze the whole genome sequence for the genetic variation and evolution.ResultsThe whole genome sequence of a G3P[8]genotype RVA strain(QD01)was obtained.The strain had a unique Wa-like genotype,G3-P[8]-I5-R1-C1-M1-A8-N1-T1-E1-H1.There were 4-13 differences in VP7 epitopes between QD01 and RVA vaccine strains RotaTeq^(TM)(G3),LLR and Rotarix.All differences showed significant variation at the key position 7-1b of the antigenic epitope.ConclusionsIn this study,the RVA strain QD01 belonged to G3-P[8]-I5-A8 genotype.Genetic evolutionary analysis hypothesized that this strain might be a novel strain generated by reassortment between porcine-and human-origin RVA strains from Asia,and QD01 had significant difference at the key positions of antigenic epitopes comparing to vaccine strains.
