摘  要:目的评价人呼吸道合胞病毒pre-F蛋白的免疫原性,为进一步的疫苗研发提供依据。方法将40只BALB/c小鼠随机分成5组,在0 d、14 d分别免疫10μg蛋白+50μg氢氧化铝、2μg蛋白+50μg氢氧化铝、10μg蛋白、2μg蛋白、50μg氢氧化铝对照,研究pre-F蛋白在小鼠体内的免疫原性和保护效果。结果所有蛋白组在免疫后28 d IgG抗体水平明显高于初次免疫。加氢氧化铝佐剂后,pre-F蛋白诱导小鼠产生的IgG抗体达到10^(4)以上。高剂量组产生的针对RSV A亚型的中和抗体平均滴度达到1000以上,针对RSV B亚型平均滴度达到600,不加铝佐剂的低剂量组小鼠中和抗体水平较低。在攻毒保护试验中,与对照组相比,蛋白免疫组的小鼠鼻洗液和肺脏病毒载量降低。结论人呼吸道合胞病毒pre-F蛋白在小鼠体内可诱导产生较高水平的中和抗体和保护效果。Objective To evaluate immunogenicity of human respiratory syncytial virus(HRSV)pre-F protein(pre-F),so as to provide the basis for further vaccine research.MethodsForty mice were divided into four groups and inoculated twice with 10μg pre-F+50μg Al(OH)_(3),2μg pre-F+50μg Al(OH)_(3),10μg pre-F,2μg pre-F and 50μg Al(OH)_(3) as the control,respectively on day 0 and day 14 to explore the in-vivo immunogenicity and protective effect of pre-F in mice.ResultsAll protein group showed that the IgG titers in 28 d after immunization were higher than that of the primary immunization.After adding Al(OH)_(3) as the adjuvant,the IgG titer induced by pre-F in mice reached more than10^(4).The average titer of neutralizing antibodies against RSV subtype A reached more than 1000 in the high-dose group,and the average titer against RSV subtype B reached 600.The level of neutralizing antibodies in the low-dose group without Al(OH)_(3)adjuvant was rather low.In the challenge protection assay,mice immunized with the pre-F had lower viral loads in nasal lotions and in lung.ConclusionsHuman respiratory syncytial virus pre-F protein can induce higher level of neutralizing antibody and protective effect in mice.
