摘 要:目的基于白纹伊蚊转录组测序结果,了解其携带蚊媒病毒情况。方法对3个白纹伊蚊野外种群和1个实验室种群构建RNA-seq文库,采用Illumina HiSeq2500j进行高通量测序,对经Trinity v2.15.1组装后的序列采用Deepvirfinder分析,鉴定其携带病毒的整体情况。结果从4个白纹伊蚊种群转录组中共鉴定到Aedes albopictus anphevirus,Aedes rhabdovirus,Longgang virus,San Gabriel mononegavirus和Yongsan tombus-like virus 5种已知RNA病毒,可能为昆虫特异性病毒,并未发现可感染脊椎动物的RNA病毒。结论本研究建立了基于白纹伊蚊转录组测序结果分析RNA病毒方法,在3个白纹伊蚊野外种群未发现已知蚊媒病毒,为进一步了解RNA病毒与伊蚊之间的动态物种特异性相互作用提供基础。Objective To understand the carriage of mosquito-borne viruses based on the results of Aedes albopictus transcriptome sequencing.MethodsRNA-seq libraries were constructed from three field populations and one laboratory population of Ae.albopictus and high-throughput sequencing was performed using Illumina HiSeq2500j system.The sequences were assembled by Trinity v2.15.1 and analyzed using Deepvirfinder to identify the overall carriage status of the viruses.ResultsA total of five known RNA viruses were identified,namely Aedes albopictus anphevirus,Aedes rhabdovirus,Longgang virus,San Gabriel mononegavirus and Yongsan tombus-like virus.The viruses may be insect-specific viruses(ISVs),and no vertebrate-infecting RNA viruses was found.ConclusionsIn this study,we established a method to analyze RNA viruses based on the transcriptome sequencing results of Ae.albopictus,and no known mosquito-borne viruses were detected in three field populations of Ae.Albopictus.This provides a basis for further understanding of the dynamic species-specific interactions between RNA viruses and Aedes mosquitoes.
