摘  要:在分子内含有五元碳-碳螺环结构的罗丹明类化合物RCK的结构中,引入次氯酸(HOCl)的识别基团N,N-二甲基硫代氨基甲酸酯,构建了具有双锁识别基团的HOCl荧光探针RCK-SCN。由于探针RCK-SCN结构中含有N,N-二甲基硫代氨基甲酸酯和α-氰基酮2个对HOCl具有识别功能的基团,因此显著提高了探针对HOCl的选择性。实验结果表明,探针RCK-SCN在578nm处的荧光强度与HOCl的浓度在0~12μmol/L范围内呈现良好的线性关系,该方法测定HOCl的检出限为31.7 nmol/L。探针RCKSCN成功用于RAW264.7细胞中外源性和内源性HOCl的荧光成像分析。A hypochlorous acid(HOCl) fluorescent probe RCK-SCN with double-locked recognition groups was constructed by introducing a classical HOCl recognition group N,N-dimethylthiocarbamate to a rhodamine derivative(RCK) containing a five-membered carbon-carbon spirocycle structure.The probe RCK-SCN showed remarkably high selectivity toward HOCl owing to having dual HOCl recognition groups of N,N-dimethylthiocarbamate and α-cyanoketone.Preliminary experimental results showed that there was a good linear relationship between the fluorescence intensity of probe RCK-SCN at 578 nm and the concentration of HOCl in the range of 0-12 μmol/L,and the detection limit for HOCl was calculated as 31.7 nmol/L.Probe RCK-SCN has been successfully applied to the fluorescent imaging of HOCl in living cells.
