摘  要:选择具有高发光强度的量子点荧光微球作为信号标记物,建立了一种快速灵敏检测喹乙醇的量子点微球免疫层析法。对探针偶联pH、碳二亚胺盐酸盐(EDC)用量、喹乙醇抗体浓度、 T线抗原浓度和探针体积进行了优化。通过量子点微球试纸条检测仪读取试纸条上检测线和质控线的荧光信号强度,以喹乙醇浓度为横坐标,以检测线(T)和质控线(C)荧光信号强度的比值(T/C)为纵坐标,建立标准曲线。结果表明,该方法定量检测范围为0.25~5μg/L,半抑制率为1.24μg/L,回收率在83.6%~118.9%之间,与卡巴氧、喹胺醇和喹烯酮等结构类似物的交叉反应率均小于0.1%。该方法适用于大批量样品的现场快速筛查。Quantum dot fluorescent nanobeads with high luminescence intensity were selected as signal markers to establish a fast and sensitive quantum dot nanobeads immunochromatography method for detecting olaquindox.The pH,carbodiimide hydrochloride(EDC) dosage,concentrations of olaquindox and T-line antigen,and probe volume were optimized.With a quantum dot strip reader,the fluorescent signal intensity of test line(T) and control line(C) on lateral flow strip was recorded.The standard curve was established by plotting the T/C ratio against the concentration of olaquindox.The results indicated that the quantitative detection range of this method was 0.25-5 μg/L,and the half inhibition rate is 1.24 μg/L.The recovery rates ranged from 83.6% to 118.9%,and the cross-reactivity with structurally similar compounds such as carbadox,kuianchun,and quinocetone were all less than 0.1%.This method is suitable for rapid on-site screening of large quantities of samples.
