摘  要:目的探讨黄连干预下雌激素水平对小鼠粪便菌群结构的影响,为黄连的药理作用及临床应用提供新的理论依据。方法选用SPF级雌性小鼠,随机分为假手术组(SHAM)、卵巢摘除手术组(OVX)、卵巢摘除手术+雌激素组(E_(2))。给予相同剂量的黄连灌胃后,采用高通量测序技术对小鼠粪便样本进行16S rRNA基因测序,观察各组小鼠粪便菌群结构的变化。结果卵巢摘除改变小鼠雌激素水平,导致OVX组小鼠的粪便菌群结构与SHAM组相比发生了显著变化(P<0.05),通过雌激素治疗,E2组与SHAM组菌群相近。结论黄连干预下雌激素水平对小鼠粪便菌群结构的影响显著,该研究为理解雌激素与肠道菌群之间的相互作用以及根据不同性别使用黄连药物提供新的线索和思路。Objective To explore the effect of Rhizoma Coptidis in intervening the estrogen levels on the structure of fecal microbiota in mice,providing new theoretical evidence for the pharmacological effects and clinical applications of Rhizoma Coptidis.Methods Specific Pathogen Free(SPF)female mice were randomly divided into the sham operation group(SHAM),the ovariectomy group(OVX),and the ovariectomy+estrogen group(E_(2)).After oral administration of the same dose of Rhizoma Coptidis,high-throughput sequencing technology was used to perform 16S rRNA gene sequencing on the fecal samples of the mice,observing changes in the fecal microbiota structure across the groups.Results Ovariectomy altered the estrogen levels in mice,resulting in significant changes in the fecal microbi⁃ota structure of the OVX group compared to the SHAM group(P<0.05).With estrogen treatment,the microbiota structure of the E2 group was similar to that of the SHAM group.Conclusion The effect of Rhizoma Coptidis in intervening the estrogen levels on the struc⁃ture of fecal microbiota in mice was significant.This study provides new insights for understanding the interaction between estrogen and gut microbiota,as well as for the use of Rhizoma Coptidis based on sex differences.
