摘 要:目的比较胡芦巴生品(R-TFG)及其酒制品(RW-TFG)的保肝作用和化学成分差异。方法采用四氯化碳(CCl4)诱导的肝损伤小鼠模型评估药效。采用超高效液相色谱四极杆飞行时间质谱(UPLC-Q-TOF/MS)对化学成分进行分析,采用正交偏最小二乘判别分析(OPLS-DA)筛选R-TFG和RW-TFG的不同化学成分,通过精确的分子量、MS/MS中的碎片离子以及与文献报道的数据做对比进行结构推断。应用相关性分析找出R-TFG和RW-TFG中与功效相关的化合物。结果RW-TFG组AST和ALP水平较低,而R-TFG组的SOD和GSH水平较高。RW-TFG中胡芦巴碱和黄酮类成分的丰度较高,甾体皂苷在酒制过程中发生了转化。相关性分析表明,胡芦巴碱与ALP相关,而黄酮类和甾体皂苷与抗氧化指数(即SOD和GSH)相关。结论酒制使胡芦巴化学成分和功效发生改变,根据相关性分析,可以找出导致药效变化的化学成分。Objective To compare the hepatoprotective effects and chemical constituents differences between raw(unprepared)Trigonel⁃la foenum-graecum L.(R-TFG)and its processed product with rice wine(RW-TFG).Methods A carbon tetrachloride(CCl4)-induced liver injury mouse model was used to evaluate the efficacy.The chemical constituents of R-TFG and RW-TFG were analyzed using UPLC-Q-TOF/MS,and the different chemical constituents of R-TFG and RW-TFG were screened by OPLS-DA.Structure inference was performed based on accurate molecular weight,fragment ions in MS/MS,and comparison with published data.Correlation analysis was used to identify the compounds associated with efficacy in both R-TFG and RW-TFG.Results The RW-TFG group ex⁃hibited lower levels of AST and ALP,while the R-TFG group showed higher levels of SOD and GSH.The abundance of trigonelline and flavonoids was higher in RW-TFG,while steroidal saponins underwent conversion during the wine processing.Correlation analysis showed that trigonelline was associated with ALP,while flavonoids and steroid saponins were associated with antioxidant indexes(SOD and GSH).Conclusion Rice wine processing altered the chemical composition and efficacy of Semen Trigonellae,and the chemical con⁃stituents responsible for these changes in efficacy can be identified through correlation analysis.
