摘 要:目的观察电针对动脉粥样硬化(AS)小鼠主动脉miRNA223-3p、NLRP3、IL-1β表达的影响,探析理中通脉法针刺改善AS的效应机制。方法SPF级ApoE^(-/-)雄性小鼠30只随机分为模型组、西药组和电针组,每组10只,高脂饲料喂养8周造模,另设C57BL/6J小鼠10只作为空白组,普通饲料喂养。治疗11周,空白组和模型组不予干预;西药组采用阿托伐他汀钙片稀释液灌胃;电针组取特定穴位施以电针。治疗期间记录小鼠体质量变化。治疗结束后HE染色观察小鼠主动脉壁组织形态;微板法检测血脂四项变化;WB检测小鼠主动脉NLRP3、IL-1β蛋白表达;RT-qPCR检测小鼠主动脉miR⁃NA223-3p、NLRP3 mRNA表达。结果与空白组对比,模型组小鼠主动脉出现明显的AS改变,体质量增加,血脂水平异常,主动脉miRNA223-3p表达降低,NLRP3、IL-1β表达升高(P<0.01);与模型组对比,西药组和电针组小鼠AS病理表现有所改善,体质量下降,血脂水平异常有所改善,主动脉miRNA223-3p表达升高,NLRP3、IL-1β表达降低(P<0.01或P<0.05)。结论理中通脉法针刺对AS病变有明显改善作用,这可能是通过miRNA223-3p/NLRP3途径改善炎症反应实现的。Objective To observe the effect of electroacupuncture(EA)on the expressions of miRNA223-3p,NLRP3 and IL-1βin the aorta of mice with atherosclerosis(AS)and to explore the mechanism of acupuncture in improving AS by Lizhong Tongmai Therapy(LZTMT).Methods Thirty SPF ApoE^(-/-)male mice were randomly divided into the model group,the Western medicine group and the EA group,with 10 mice in each group.Mice were fed with a high-fat diet for 8 weeks to establish the model.Ten C57BL/6J mice were set as the blank group and fed with ordinary diet.The blank group and the model group were not intervened for 11 weeks;the West⁃ern medicine group was gavaged with atorvastatin calcium tablet dilution,and the EA group was electroacupunctured at specific acu⁃points.The changes in the body weight of the mice were recorded during the treatment.After the treatment,the morphology of the aortic walls of mice was observed by HE staining;the changes in four blood lipids were detected by microplate method;the expressions of NL⁃RP3 and IL-1βproteins in the aorta of mice were detected by Western blot(WB);and the expressions of miRNA223-3p and NLRP3 mRNA in the aorta of mice were detected by RT-qPCR.Results Compared with the blank group,the model group mice showed signifi⁃cant AS changes in the aorta,increased body weight,abnormal blood lipid levels,decreased expression of aortic miRNA223-3p,and increased expression of NLRP3 and IL-1β(P<0.01);compared with the model group,the AS pathological manifestations of mice in the Western medicine group and EA group were alleviated,body weight was decreased,abnormal blood lipid levels were improved,aortic miRNA223-3p expression was increased,and NLRP3 and IL-1βexpressions were decreased(P<0.01 or P<0.05).Conclusion⁃Acupuncture with LZTMT has a significant healing effect on AS lesions,which may be achieved by improving the inflammatory response through the miRNA223-3p/NLRP3 pathway.
