摘  要:目的:探讨萝卜硫素(SFN)对妊娠高血压(PIH)大鼠血管内皮功能的影响及作用机制。方法:选取无特定病原体(SPF)级怀孕SD雌性大鼠30只,随机分为对照组、PIH组、SFN低剂量组(15 mg/kg)、SFN高剂量组(30 mg/kg)和LY294002组(30 mg/kg SFN+5 mg/kg LY294002)。构建PIH大鼠模型,测定大鼠收缩压;二喹啉甲酸(BCA)法测定尿蛋白含量;酶联免疫吸附法(ELISA)检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、血管内皮生长因子(VEGF)、血管细胞黏附分子1(VCAM-1)、内皮素-1(ET-1)、一氧化氮(NO);蛋白免疫印迹法(Western Blot)检测胎盘组织磷酸肌醇3-激酶(PI3K)、磷酸化PI3K(p-PI3K)、蛋白激酶B(AKT)、磷酸化AKT(p-AKT)、VEGF蛋白表达水平。结果:与对照组相比,PIH组收缩压、尿蛋白含量、TNF-α、IL-1β、IL-6、MDA、ET-1、VCAM-1水平明显升高(P<0.05),血清VEGF、NO、SOD、GSH及p-PI3K/PI3K、p-AKT/AKT、VEGF蛋白水平明显下降(P<0.05);与PIH组相比,SFN低剂量组、高剂量组收缩压、尿蛋白含量以及TNF-α、IL-1β、IL-6、MDA、ET-1、VCAM-1水平明显降低(P<0.05),血清VEGF、NO、SOD、GSH以及p-PI3K/PI3K、p-AKT/AKT、VEGF蛋白水平明显升高(P<0.05);PI3K抑制剂LY294002可减弱SFN对PIH大鼠血管内皮功能的保护作用。结论:SFN对PIH大鼠血管内皮功能有保护作用,可能与激活PI3K/AKT/VEGF信号通路有关。ObjectiveObjective:To explore the effect of sulforaphane(SFN)on vascular endothelial function in pregnancy-induced hypertension(PIH)rats.MethodsMethods:Thirty pregnant SD rats free of specific pathogen(SPF)were randomly divided into control group,PIH group,SFN low-dose group(15 mg/kg),SFN high-dose group(30 mg/kg),and LY294002 group(30 mg/kg SFN+5 mg/kg LY294002).PIH rat model was established and systolic blood pressure was measured.Bicinchoninic acid(BCA)method was used to determine the urine protein content.Enzyme-linked immunosorbent assay(ELISA)was used to measure serum tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione(GSH),vascular endothelial growth factor(VEGF),vascular cell adhesion molecule 1(VCAM-1),endothelin-1(ET-1),and nitric oxide(NO).Western Blot was used to detect protein expression levels of phosphoinositide 3-kinase(PI3K),phosphorylated PI3K(p-PI3K),protein kinase B(AKT),phosphorylated AKT(p-AKT),and vascular endothelial growth factor(VEGF)in placenta tissue.ResultsResults:Compared with the control group,the systolic pressure,urine protein content,levels of TNF-α,IL-1β,IL-6,MDA,ET-1,and VCAM-1 of the rats in the PIH group increased(P<0.05),the protein levels of VEGF,NO,SOD,GSH,p-PI3K/PI3K,p-AKT/AKT,and VEGF decreased(P<0.05).Compared with the PIH group,the systolic pressure,urine protein content,levels of TNF-α,IL-1β,IL-6,MDA,ET-1,and VCAM-1 of rats in the SFN low and high dose groups decreased(P<0.05),the protein levels of VEGF,NO,SOD,GSH,p-PI3K/PI3K,p-AKT/AKT,and VEGF increased(P<0.05).The PI3K inhibitor LY294002 attenuated the protective effect of SFN on vascular endothelial function in PIH rats.ConclusionConclusion:SFN shows a protective effect on vascular endothelial function in PIH rats,which may be related to the activation of PI3K/AKT/VEGF signaling pathway.
