摘  要:目的 通过两样本孟德尔随机分析探究基因决定的儿童肥胖与注意力缺陷多动障碍之间的因果关系。方法 从全基因组关联研究汇总数据集中获取儿童肥胖的数据,筛出4个与儿童肥胖相关的单核苷酸多态性作为工具变量,使用逆方差加权法、加权中位数法和MR Egger等多种方法进行孟德尔随机化分析探究儿童肥胖与注意力缺陷多动障碍之间的因果关系,其中逆方差加权法为主要方法。结果 逆方差加权法结果表明,儿童肥胖与注意力缺陷多动障碍风险呈正相关(OR=1.126;95%CI:1.022~1.241,P=0.010),使用加权中位数法观察到类似的结果(OR=1.133;95%CI:1.011~1.270,P=0.030),敏感性分析结果显示没有水平多效性和异质性。结论 基因决定的儿童肥胖与注意力缺陷多动障碍风险之间具有因果关系。Objective To explore the causal relationship between genetically determined childhood obesity and attention-deficit/hyperactivity disorder(ADHD)using two-sample Mendelian randomization analysis.Methods Data on childhood obesity were obtained from a genome-wide association study(GWAS)summary dataset.Four single nucleotide polymorphisms(SNPs)associated with childhood obesity were selected as instrumental variables.Various methods,including inverse variance weighted,weighted median,and MR Egger,were employed to conduct Mendelian randomization analysis to investigate the causal relationship between childhood obesity and ADHD,with inverse variance weighted being the primary method.Results The results from inverse variance weighted analysis indicated a positive correlation between childhood obesity and the risk of ADHD(OR=1.126;95%CI:1.022~1.241,P=0.010).Similar results were observed using the weighted median method(OR=1.133;95%CI:1.011~1.270,P=0.030).Sensitivity analysis showed no horizontal pleiotropy or heterogeneity.Conclusion There is a causal relationship between genetically determined childhood obesity and the risk of attention deficit hyperactivity disorder.
