摘 要:为探索西藏小型猪作为猪链球菌2型、7型、9型感染模型的可行性,本研究用2型、7型、9型猪链球菌分别以2×10^(8)、7×10^(9)、2×10^(9)CFU的剂量颈部肌肉接种60日龄的西藏小型猪,对照组猪同样方式接种等体积生理盐水,每组5头仔猪。接种后定期监测仔猪的临床症状和血液中带菌情况,30 d后安乐死全部组别仔猪,观察其组织病理学变化。结果显示:与对照组相比,攻毒组仔猪均表现出发热、食欲减退、精神沉郁等临床症状;攻毒后仔猪血液带菌的时间为3~6 d,组织病理学变化结果显示2型猪链球菌对西藏小型猪的致病力最强,其次为7型,病变主要集中在肺脏,脑、肝脏或关节等部位也有不同程度的病变。本研究初步确定西藏小型猪可作为猪链球菌2型、7型、9型人工感染模型,可为临床猪链球菌的致病机制研究和动物模型建立提供参考依据。To explore the feasibility of Tibetan miniature pigs as the infection model of serotypes 2,7 and 9 Streptococcus suis(SS),each of these serotypes were inoculated into 5 sixty-days-old healthy pigs at doses of 2×10^(8),7×10^(9),2×10^(9)CFU or saline by intramuscular injection in necks.Other 5 control pigs were injected with equal volume of saline.The clinical signs and bacteria loads in the blood samples of these piglets were monitored regularly after inoculation,and all pigs were euthanized 30 days later to examine the histopathological changes.The results showed that the significant clinical signs were observed in all pigs infected with SS serotypes 2,7 and 9 strains,such as fever,loss of appetite and depression.The bacteria were detected in blood samples from 3 to 6 days post inoculation.The results of histopathological changes indicated SS serotype 2 strain was most severe followed by serotype 7.The lesions were mainly concentrated in lungs,and some lesions were observed in brains,livers and joints.The results of this preliminary study indicated that Tibetan miniature pigs could be used as the experimental infection models of SS serotypes 2,7 and 9,which provided a reference for further research of pathogenic mechanism animal models.
