长角血蜱铁蛋白HlFer1参与唾液腺退化研究

摘  要:本研究旨在研究长角血蜱(H.longicornis)铁蛋白1(Ferritin1)与蜱唾液腺退化的关系,为唾液腺退化机制研究提供理论基础。利用PCR方法扩增长角血蜱Ⅰ型铁蛋白(HlFer1)基因,构建重组表达质粒,表达蛋白后制备多克隆抗体用于蛋白水平验证。收集不同吸血时期的长角血蜱唾液腺,经qPCR分析HlFer1分子的转录水平以及Western blot验证蛋白表达水平,发现HlFer1基因转录水平和蛋白表达水平均随唾液腺退化进程呈递增趋势。再利用RNAi技术干扰HlFer1基因,发现实验组蜱的线粒体出现膜密度增高,嵴减少、体积缩小等特征,唾液腺退化加快,表明HlFer1参与蜱唾液腺退化过程,可能与铁死亡相关。该研究结果可为蜱的生物防控奠定基础。The study aimed to investigate the relationship between Ferritin1 and salivary gland degeneration in ticks and to provide a theoretical basis for studying the mechanism of salivary gland degeneration.PCR was used to amplify the Haemaphysalis longicornis type I ferritin(HlFer1)gene,recombinant expression plasmid was constructed,and polyclonal antibody was prepared for protein level verification after protein expression.Salivary glands of H.longicornis from different periods of bloodsucking were collected and analyzed for transcript levels of HlFer1 molecules by qPCR and protein expression levels were verified by Western blot,and found to exhibit increased levels of the HlFer1 gene.The transcriptional and protein expression levels of the HlFer1 gene both showed a progressively increasing trend with the degeneration of the salivary glands.Ferritin1 was then interfered with using RNAi technology.It was found that mitochondria in the experimental group showed features such as increased membrane density,decreased cristae,and reduced volume,suggesting that HlFer1 is involved in the process of tick salivary gland degeneration,possibly associated with iron death,laying the foundation for the biological control of ticks.
