摘 要:鸭坦布苏病毒病是由坦布苏病毒(DTMUV)感染引起的一种可以引起蛋鸭产蛋率严重下降、雏鸭出现神经症状甚至死亡的传染病。目前该病在免疫防控等方面取得了较大的研究进展,但是宿主与DTMUV感染之间的免疫机制还有待我们进一步研究。本研究应用DTMUV感染鸭胚成纤维细胞(DEF),利用荧光定量PCR(qPCR)检测抗病毒信号通路主要细胞因子(RIG-1、IRF-7、STAT1、IFN-α、IFN-β、IFN-γ)、主要组织相容性复合体(MHCⅠ、MHCⅡ)、白细胞介素(IL-1β、IL-6、IL-8)、趋化炎性因子(CCL-4、CCL-5)和凋亡相关因子(Fas、BCL2、BCL2A、APAF-1、CASP7、BiD、MCL、FADD)mRNA转录水平。结果显示DTMUV感染DEF 12 h后,有少数细胞因子呈现轻微下调,但在感染24 h后所测的21种细胞因子全部呈现上调,其中IRF7、IFN-β、MHCII、IL-1β、CCL4与对照组相比上调了4.81倍、14.58倍、3.24倍、4.68倍、11.73倍。对TMUV感染DEF后免疫相关因子mRNA转录水平的变化分析发现,病毒感染DEF细胞24 h后,细胞产生明显的炎性反应,同时激活抗病毒信号通路、细胞凋亡通路,促进多种免疫相关因子的表达。该研究为DTMUV感染宿主致病机制与宿主抗病毒固有免疫反应的研究奠定了良好的基础。Duck tembusu virus(DTMUV)is a kind of infectious disease that causes serious decrease of egg production and nervous system signs in laying ducks.Although significant research progress has been made in the field of prevention and control,the immune mechanism between host and DTMUV infection still needs to be further studied.In this study,DTMUV was used to infect duck embryos fibroblasts(DEF)and fl uorescence quantitative PCR(qPCR)was used to detect mRNA transcription levels of major cytokines(RIG-1,IRF-7,STAT1,IFN-α,IFN-β,and IFN-γ),major histocompatibility complex(MHCⅠand MHCⅡ),interleukin(IL-1,IL-6 and IL-8),chemotactic infl ammatory factors(CCL-4 and CCL-5)and apoptosis-related factors(Fas,BCL2,BCL2A,APAF-1,CASP7,BiD,MCL and FADD)in the antiviral signaling pathway.DEF cells were infected with DTMUV for 12 hours,a few cytokines showed a slight downregulation of a few cytokines in DEF cells at 12 h post infection and up-regulation of all 21 cytokines at 24 h.The transcriptions were respectively up-regulated 4.81,14.58,3.24,4.68 and 11.73 times for IRF7,IFN-β,MHCII,IL-1β,CCL4 as compared with the control group.The mRNA and transcript levels of immune-related factors were also analyzed after TMUV infection of DEF cells.At 24 h post infection,the cells produced obvious infl ammatory response,activated antiviral signaling pathway and apoptosis pathway and promoted the expression of multiple immune-related factors.In conclusion,this study laid a foundation for the further study of the pathogenesis of DTMUV infection and antiviral innate immune response of the hosts.
