摘  要:为了研究缺失RLORF6、Meq基因的鸡马立克病病毒(MDV)MDLV21株对SPF鸡的致病性与免疫保护效果。通过接种SPF鸡,比较MDLV21株对鸡的体重、免疫器官指数、灭活疫苗诱导抗体水平的影响并统计死亡率和肿瘤率;通过攻毒保护试验,统计分析MDLV21株与CVI988/Rispens株对MDV超强毒株Md5的免疫保护指数。结果显示:MDLV21株接种SPF鸡的体重、胸腺和法氏囊免疫器官指数均与对照组SPF鸡差异不显著;接种鸡与对照组鸡的禽流感、新城疫抗体滴度相似。MDLV21株接种鸡在整个周期没有死亡以及肿瘤发生,脾脏、肝脏组织切片显示细胞结构正常。MDLV21株、CVI988/Rispens株对感染Md5的SPF鸡分别提供90%、70%的免疫保护。综上,MDV MDLV21株对SPF鸡没有致病性,对MDV Md5株具有良好的免疫保护。本研究可为MD的防控提供1株新的生物安全性、免疫效果良好的疫苗候选株。Safety and immune protection of Marek’s disease virus(MDV)strain MDLV21 with deletion of RLORF6 and Meq genes were evaluated in specific-pathogen-free(SPF)chickens.The SPF chickens were inoculated with the inactivated vaccine MDLV21 strain.The vaccinated chickens were observed for body weight and mortality and examined for immune organ index,antibody level and tumor formation.Subsequently,these vaccinated chickens were challenged with the very virulent MDV strain Md5 to evaluate the immune protection of MDLV21 and CVI988/Rispens.The results showed that the body weights,immune organ indexes of thymuses and bursas of fabricius of SPF chickens inoculated with MDLV21 strain were not significantly different from those of control chickens.The AIV and NDV antibody titers of the MDLV21 inoculated chickens were similar to the controls.Moreover,the MDLV21 inoculated chickens did not die or develop tumors during the whole experimental period and tissue sections of spleens and livers showed normal cell structures.The MDLV21 and CVI988/Rispens provided 90%and 70%protection for SPF chickens against Md5 challenge,respectively.Collectively,MDV MDLV21 was safe to SPF chickens and provided good immune protection against MDV Md5 challenge.The study provided a new vaccine candidate strain with good safety and immune protection for MDV infection.
