摘  要:本研究采集疑似患有牛支原体肺炎的病变肺脏进行牛支原体的分离鉴定。通过菌落形态观察、PCR鉴定、序列分析等方法进行鉴定,结果表明成功分离到1株牛支原体,命名为YJ-22株。生化特性研究发现:分离株不能利用葡萄糖、精氨酸、明胶、尿素、甘露醇等营养物质,但可以利用乳糖,与牛支原体生化特性相符。生长曲线测定结果显示,牛支原体YJ-22株在37℃培养9 h可达对数生长期,在18~45 h处于平台期。使用牛支原体YJ-22株对3月龄犊牛进行气管注射攻毒,可产生典型的牛支原体肺炎症状和病变,说明该分离株可以感染犊牛。使用牛支原体YJ-22株制备灭活疫苗免疫兔子后产生的IgG抗体效价可以达到1∶128,说明该分离株有较好的免疫原性。本研究分离得到1株牛支原体,该株牛支原体对犊牛具有较强的致病力,且具有良好的免疫原性,可以作为牛支原体疫苗候选株。In this study,a lung sample was collected from a diseased cattle suspected of having Mycoplasma bovis pneumonia were collected for mycoplasma isolation and identification.The isolated mycoplasma strain was identified by colony morphology observation,PCR,sequence analysis,etc.The results showed that one M.bovis strain was isolated and designated as YJ-22 strain.The biochemical study found that the YJ-22 strain did not use nutrients such as glucose,arginine,gelatin,urea,mannitol,etc.but used lactose,which was consistent with M.bovis.The YJ-22 strain was cultured at 37℃for 9 hours to reach the logarithmic growth and 18-45 hours to the plateau stage.Two-month-old calves infected with the YJ-22 strain via tracheal injection produced typical signs and lesions of M.bovis pneumonia,indicating that the YJ-22 strain was pathogenic to cattle.The IgG titer in rabbits immunized with the inactivated YJ-22 strain reached 1:128,indicating that this train induced good antibody response.These results showed that the M.bovis strain YJ-22 isolated from a disease cattle had strong pathogenicity to calves and produced good antibody response in rabbits,which could be used as a vaccine candidate for M.bovis.
