摘  要:猪巨细胞病毒(PCMV)感染可导致猪食欲废绝,母猪流产,产木乃伊胎,仔猪发育不良等临床症状,且分布广泛,对异种器官移植也存在潜在威胁。本研究针对PCMV保守基因DPOL设计引物探针,建立TaqMan实时荧光定量PCR方法。结果显示:检测限可达10拷贝,灵敏度高;变异系数<2%,标准曲线R2=0.998,重复性好,具有高特异性;对上海市及江苏省常州市170份血清样本进行检测,5份样品为可疑结果。本研究建立的方法可用于养殖场猪感染PCMV的筛查,对该病防控具有重要意义。Porcine cytomegalovirus(PCMV)infection in pigs causes clinical sign such,as loss of appetite,miscarriage of sows,production of mummified fetuses,and poor development of piglets.The PCMV infection is widely distributed and also poses a potential threat to xenotransplantation.In this study,a primer probe was designed for the PCMV conservative gene DPOL for development of a TaqMan real-time fluorescence quantitative PCR method.The PCR method showed high sensitivity,good repeatability,and high specificity with the detection limit up to 10 copies,the coefficient of variation was less than 1.41%,and the standard curve R 2 at 0.998.A total of 170 serum samples from Shanghai and Changzhou,Jiangsu were tested and 5 samples were found suspicious.This method could be used for screening PCMV infection on pig farms,and was of great significance for the prevention and control of the disease.
