摘 要:为了解四川省地区猪圆环病毒3型(PCV3)流行毒株的分子及遗传变异规律,试验采用生物学软件,设计3对特异性引物对从四川地区部分养殖场猪群中收集的15份PCV3阳性样品进行分段扩增、全基因序列遗传变异分析。结果表明:PCR分段扩增分别得到大小为623、803和753 bp的条带,阳性克隆菌测序结果与GenBank中PCV3的同源性均大于98.6%;15株样品毒株的全基因核苷酸序列的同源性为98.1%~99.8%,推导氨基酸序列的同源性为95.8%~99.2%,由Cap基因的遗传进化分析发现从四川地区获取的15株PCV3毒株属于3a和3b两个亚群。说明四川地区流行的PCV3毒株至少存在3a和3b 2个亚群,该亚群的变异程度低、较为保守。In order to understand the molecular and genetic variation of Porcine circovirus type 3(PCV3)strains prevalent in Sichuan province,this experiment used bioinformatics software to designed three pairs of primers to amplify the genomes of PCV3 and performed the genomic sequencing and genetic variation analysis on 15 PCV3 strains from pig herds of some large-scale pig farms.The results showed that three bands with sizes of 623803 and 753 bp were obtained by PCR amplification and the sequencing analysis of the positive clones revealed more than 98.6%homologous to the PCV3 in GenBank.The homology of the whole genome nucleotide sequences and deduced amino acid sequences of these 15 strains were 98.1%-99.8%,95.8%-99.2%,respectively.Phylogenetic analysis based on the cap gene showed that these 15 strains belonged to PCV3a and PCV3b subgroups,suggesting epidemic occurrence of PCV3a and PCV3b two subgroups in Sichuan province.These two subgroups were relatively conserved with low variation.
