1株鸽新城疫病毒分离鉴定与全基因组序列分析

摘  要:本研究对2020年从广西地区采集的疑似鸽新城疫临床样品,进行了病毒分离、致病力鉴定,并进行全基因组测序和遗传进化分析。结果成功分离到1株鸽新城疫病毒,致病力试验证实为高毒力毒株;该毒株F蛋白裂解位点序列为112RRQKRF117。通过绘制F基因和全长基因组遗传进化树,确定该毒株属Ⅵ.2.1.1.2.2Ⅵk型,与流行毒株Pi/CH/TJ2017亲缘关系最近,与新城疫疫苗株La Sota遗传关系较远;蛋白序列分析结果显示,F蛋白的两个高度疏水区信号肽区(1-31)和融合肽区(117-141)共存在13个氨基酸位点变异;HN蛋白中和表位共有6个氨基酸位点的突变。上述突变可能造成该分离株的F蛋白和HN蛋白抗原性及疏水性有一定程度变化,影响La Sota疫苗株的保护效力,导致鸽新城疫在鸽群中暴发。In this study,virus isolation,pathogenicity testing,whole genomic sequencing and genetic evolution analysis were performed on suspected pigeon Newcastle disease clinical samples collected in 2020 from Guangxi.A Highly pathogenic pigeon paramyxovirus 1 with the F protein cleavage site sequence of 112RRQKRF117 was isolated and identified from the tested samples.This strain was identified as GenotypeⅥ.2.1.1.2.2Ⅵk by mapping the F gene and full-length genomic evolutionary tree,which was the most closely related to the prevalent strain Pi/CH/TJ2017 and genetically distant from the commonly used Newcastle disease vaccine strain La Sota.The protein sequence analysis showed that there were 13 amino acid site variations in the signal peptide region(1-31)and the fusion peptide region(117-141)of the two highly hydrophobic regions of the F protein and 6 amino acid site mutations in the neutralization epitope of the HN protein.These mutations mingt have caused some changes in the antigenicity and hydrophobicity of the F and HN proteins of this isolate,affecting the protection F and HN of the La Sota vaccine strain and leading to an outbreak of Newcastle disease in the pigeon population.
