摘  要:磷酸奥司他韦(OP)是一种神经氨酸酶抑制剂,临床上用于治疗A和B型流感病毒。实验通过在最佳聚合条件下,在玻碳电极(GCE)上电化学聚合纳米金(AuNPs)/聚L-鸟氨酸(PL-Orn),制备了一种用于测定OP的高灵敏度复合传感器(AuNPs/PL-Orn/GCE)。采用循环伏安法(CV)和扫描电镜(SEM)对该电极进行了表征:PL-Orn在GCE表面形成了一层均匀的膜,AuNPs在PL-Orn均匀地分散,呈针簇状。在最佳测定条件下,该修饰电极可用于OP的测定,线性范围为8.00×10^(-8)~2.00×10^(-5)mol/L,相关系数R为0.9941,检出限为4.00×10^(-8)mol/L。把该电极用于OP胶囊样品的测定,结果满意。利用电化学方法和紫外可见光谱研究了OP与DNA的相互作用,结果表明:OP与DNA发生了增色效应。Oseltamivir phosphate(OP),a neuraminidase inhibitor,is used clinically to treat influenza A and influenza B.A sensor of poly(AuNPs)/poly(L-Ornithine)gold nanoparticles modified glassy carbon electrode,AuNPs/PL-Orn/GCE for determination of OP is fabricated by electropolymerizing AuNPs and PL-Orn on a GCE under optimal polymerization conditions.The electrode is characterized by cyclic voltammetry(CV)and scanning electron microscopy(SEM).The images show that the PL-Orn formed a layer of uniform membrane,and the deposited AuNPs are uniformly dispersed on the PL-Orn/GCE,which is given shape in the form of lusters of needle.The electrochemical experiments show that,under the optimal conditions,the modified electrode can be used for determination of OP.The linear range of 8.00×10^(-8)~2.00×10^(-5)mol/L,with a correlation coefficient R of 0.9941 and a detection limit of 4.00×10^(-8)mol/L is obtained.The practical application of AuNPs/PL-Orn/GCE is demonstrated by the determination of OP in OP capsules samples.This electrochemical method and UV-vis spectrum are employed to investigate the interaction between OP with DNA.The results show that OP and DNA have hyperchromic effect.
