摘 要:为分析间断双线绦虫(Digramma interrupta)的线粒体基因(mtDNA)序列的遗传多态性,探讨其作为分子标记的应用潜力,于云南省迪庆州获得的短须裂腹鱼体内分离出21条间断双线绦虫,分别提取基因组DNA,扩增ATP酶亚基6(p atp 6)、烟酰胺腺嘌呤二核苷酸脱氢酶亚基5(p nad 5)、核糖体RNA小亚基(p rrn S)部分基因,并进行序列分析。基于这3个基因,以多房棘球绦虫为外群,采用贝叶斯法构建进化树,分析间断双线绦虫与其他绦虫之间的系统发育关系。结果发现,p atp 6、p nad 5和p rrn S扩增产物的长度分别为543 bp、628 bp和1133 bp,种内差异依次为0~1.1%、0~1.4%和0~0.4%,在进化树上与间断双线绦虫与舌状绦虫亲缘关系最近。结果表明,3个mtDNA序列变异程度从低到高依次为p rrn S<p atp 6<p nad 5,p nad 5是研究间断双线绦虫遗传多样性的理想线粒体分子标记。To analyze the genetic polymorphisms of genes in mitochondrial DNA(mtDNA)in Digramma interrupta,and explore their application potential as molecular markers,in this study,21 D.interrupta were isolated from Schizothorax wangchiachii,which was obtained in Diqing prefecture,Yunnan province.Genomic DNA was extracted,and paritial ATP synthase subunit 6(p atp 6),nicotinamide adenine dinucleotide dehydrogenase subunit 5(p nad 5),small subunit of ribosomal RNA(p rrn S)of D.interrupta were amplified and sequenced.A phylogenetic tree based on nucleotide sequences p atp 6,p nad 5 and p rrn S was constructed using Bayesian method,choosing Echinococcus multilocularis as an outgroup.The results found that the sizes of p atp 6,p nad 5 and p rrn S were approximately 543 bp,628 bp and 1133 bp,respectively.Intraspecific variation in p atp 6,p nad 5 and p rrn S were 0-1.1%,0-1.4%and 0-0.4%,respectively.D.interrupta and L.intestinalis were most related based on the present phylogenetic analysis.The intraspecific variation of sequences among three mt gene of D.interrupta were as follows:p rrn S<p atp 6<p nad 5.The results of present study also indicated that p nad 5 is an ideal mt molecular marker in studying the genetic diversity of species in genus Digramma.
