摘 要:为了解gga-miR-27b-3p在鸡脂肪沉积中的作用,试验以矮小品系S3和隐性白羽肉鸡为试验素材,荧光定量PCR检测gga-miR-27b-3p在腹脂、肝脏及脂肪细胞中的表达,利用生物信息学分析筛选关键靶基因。结果显示:(1)gga-miR-27b-3p在腹脂、肝脏及脂肪细胞中均有表达,且存在显著的组织和品种差异性(P<0.05)。在腹脂上,gga-miR-27b-3p在0周龄表达有品种差异(P<0.01),S3系在0周龄时显著高于14周龄(P<0.05)和16周龄(P<0.01),隐性白羽肉鸡在0周龄时极显著高于其他周龄(P<0.01);在肝脏上,gga-miR-27b-3p在16周龄有品种差异(P<0.01),S3系在16周龄显著高于其他周龄(P<0.01),隐性白羽肉鸡在16周龄显著高于0周龄(P<0.01)和8周龄(P<0.05)。(2)gga-miR-27b-3p腹脂脂肪细胞分化1 d的表达显著低于分化4 d和6 d(P<0.05);(3)KEGG及蛋白互作分析揭示GRB2、PIK3R3、PDGFRA、ERBB4、MET、KRAS和HGF是gga-miR-27b-3p的潜在靶基因,其中GRB2和PIK3R3尤为关键。Q-PCR结果表明,GRB2基因在腹脂脂肪细胞增殖期的表达量显著高于分化期(P<0.01),PIK3R3基因在分化1 d的表达量显著高于分化4 d和6 d(P<0.01)。综上所述,gga-miR-27b-3p的表达存在显著的品种和组织差异,并可能是通过靶向GRB2和PIK3R3影响脂肪沉积,结果为深入探讨gga-miR-27b-3p的功能和机制提供理论依据和数据支持。In order to understand the role of gga-miR-27b-3p in chicken fat deposition,the expression of gga-miR-27b-3p in abdominal fat,liver and adipocytes was detected by fluorescence quantitative PCR using dwarf strain S3 and cryptic white-feathered chickens as test materials,and key target genes were screened using bioinformatics analysis.The results showed that:①gga-miR-27b-3p was expressed in abdominal fat,liver and adipocytes with significant tissue and species variability(P<0.05).In ventral fat,gga-miR-27b-3p was expressed at 0 weeks of age with breed differences(P<0.01),significantly higher in S3 line at 0 weeks of age than that at 14 weeks of age(P<0.05)and 16 weeks of age(P<0.01).The expression level of cryptic white-feathered broiler chickens at 0 weeks of age was extremely significantly higher than that at any other weeks of age(P<0.01);And in the liver,gga-miR-27b-3p at 16 weeks of age There were breed differences(P<0.01),S3 line at 16 weeks of age was significantly higher than that other weeks of age(P<0.01),and cryptic white-feathered broilers were significantly higher at 16 weeks of age than that at 0 weeks of age(P<0.01)and at 8 weeks of age(P<0.05).②The expression of gga-miR-27b-3p ventral adipocytes at 1 d of dif-ferentiation was significantly lower than that at 4 d of differentiation and at 6 d of differentiation(P<0.05).③KEGG and protein interactions analysis revealed that GRB2,PIK3R3,PDGFRA,ERBB4,MET,KRAS and HGF were potential target genes of gga-miR-27b-3p,among which GRB2 and PIK3R3 were particularly critical.Q-PCR results showed that the expression of GRB2 gene was significantly higher in the proliferation phase of abdominal fat adipocytes than that in the differentiation phase(P<0.01),and the expression of PIK3R3 gene at 1 d of differentiation was significantly higher than that at 4 d and 6 d of differentiation(P<0.01).In summary,the expression of gga-miR-27b-3p had significant species and tissue differences and might affect fat depo-sition by targeting GRB2 and PIK3R3,which provided theor
