应用转移因子降低鸡传染性贫血病毒感染对SPF鸡的致病性研究

摘  要:为探索转移因子的免疫增强作用及其对鸡传染性贫血病毒(Chicken infectious anemia virus,CIAV)感染的抑制作用,试验研究了高、低两种剂量转移因子对SPF鸡CD4^(+)和CD8^(+)T淋巴细胞水平,鸡白细胞介素-12(IL-12)、γ-干扰素(IFN-γ)等6种细胞因子和Mx抗病毒蛋白表达的影响,以及对SPF鸡体重、器官发育指数、红细胞压积(HCT)值和血液、肝脏、胸腺中CIAV增殖的影响。结果显示:转移因子可引起CD4^(+)和CD8^(+)T细胞比重显著增加并上调IFN-γ、Mx蛋白表达量(P<0.01);CIAV感染组的脾脏发育指数相对于空白对照组显著增加(P<0.05),胸腺发育指数极显著降低(P<0.01),转移因子可以缓解感染CIAV带来的脾脏肿大和胸腺萎缩;转移因子可显著抑制CIAV在血液、肝脏和胸腺中的复制(P<0.01),且高剂量组抑制作用强于低剂量组;CIAV感染组血浆HCT值极显著低于空白对照组(P<0.01),高剂量转移因子使用后缓解了CIAV感染引起的HCT值下降。研究表明,转移因子对缓解CIAV感染引起的危害具有良好的应用价值,为防控鸡传染性贫血提供新的辅助措施。To explore the immune enhancing effect of transfer factor and their inhibitory effect on chicken infectious anemia virus(CIAV)infection,the experiment study the effects of high-and low-dose transfer factor on CD4^(+)and CD8^(+)T lymphocyte level,as well as the expression of 6 kinds of cytokines including interleukin-12(IL-12)andγ-interferon(IFN-γ),and Mx antiviral protein in SPF chickens.And turthermore the effects of the two doses of transfer factor on SPF chickens′body weight,organ development index,hematokrit(HCT)value,and CIAV proliferation in blood,liver and thymus were conducted.The results showed that transfer factor could significantly increase the proportion of CD4^(+)and CD8^(+)T cells and upregulate the expression of IFN-γand Mx proteins(P<0.01);The spleen development index of CIAV infected group significantly increased compared to the blank control group(P<0.05),while the thymus development index significantly decreased(P<0.01).Transfer factor could alleviate the spleen enlargement and thymus atrophy caused by CIAV infection;The transfer factor significantly inhibited the replication of CIAV in blood,liver and thymus(P<0.01),and the inhibitory effect of the high-dose group was stronger than that of the low-dose group;The plasma HCT value in CIAV infection group was significantly lower than that in the blank control group(P<0.01),and the use of high-dose transfer factor alleviated the decrease in HCT value caused by CIAV infection.The results suggested that transfer facxtor had good applicationvalue in alleviating the harm caused by CIAV infection,which proposed a new auxiliary measure for prevention and control of chicken infectious anemia.
