摘 要:为了解鹅源A型产气荚膜梭菌(Clostridium perfringens,Cp)对鹅的致病性,研究通过鸡胚致病性试验、不同感染途径攻毒试验筛选毒力较强的菌株及适合感染途径,对35日龄雏鹅进行人工致病试验,通过评估临床症状、体重变化、肠道表征评分、组织病理学变化、肠道绒毛高度及隐窝深度等研究其对鹅的致病性。结果显示:Cp-07株对鸡胚的致病性较强,鸡胚半数致死量(ELD_(50))为10^(-8.75)/0.2 mL;灌服Cp-07株可对雏鹅致死,肠道能分离到Cp,而皮下注射和静脉注射仅在第6天肠道分离到Cp;感染组雏鹅表现出精神沉郁、拉血便等症状,从第3天开始感染组体重低于对照组,肠道出现明显的肠壁变薄、出血,肠黏膜脱落并出现坏死灶,肠道表征评分高达4分;感染组肠绒毛脱落、断裂,大量红细胞、炎性细胞浸润,十二指肠及回肠固有形态消失,小肠绒毛高度不同程度下降,隐窝深度增加。研究表明,试验筛选出1株高毒力Cp,适合接种途径为灌服,单独感染Cp可引起鹅肠道损伤。To determine the pathogenicity of goose-derived Clostridium perfringens type A(Cp)in goose,chicken embryos pathogenicity test and challenge test through different infection routes were conducted in this experiment,which was used to screen the optimal virulent strain and the infection route for the artificial infection on 35-day-age goslings.The pathogenicity was evaluated by clinical symptoms,body weigh dynamics,intestinal lesions score,histopathological changes and intestinal villus height and crypt depth.The results showed that Cp-07 strain had the higher pathogenicity with median embryo lethal dose(ELD_(50))of 10^(-8.75)/0.2 mL.The oral gavage led to lethal infections in goslings,and Cp was isolated from the intestines,while subcutaneous and intravenous injections resulted in the isolation of Cp from the intestines only on the 6th day.The goslings in the infected group displayed symptoms such as lethargy and bloody diarrhea,with body weight lower than that of the control group starting from the third day.In the infected goslings,there were intestinal wall thinning,hemorrhage,mucosal sloughing and necrotic lesions,with intestinal lesion scores reaching up to 4 points,and the intestine had sloughed and fragmented intestinal villi,significant infiltra-tion of erythrocytes and inflammatory cells,and the loss of normal architecture in the duodenum and ileum.Additionally,a reduc-tion in villus height and an increase in crypt depth were observed in the small intestine.The results suggested that highly virulent Cp was successfully isolated and oral gavage was the suitable infection route,and Cp infection alone could cause intestinal dam-age in goose.
