盐炙前后知母对阴虚便秘大鼠润肠通便作用影响及机制探讨

摘  要:本文研究盐炙前后知母对阴虚便秘大鼠的影响,探讨其炮制后是否能够通过调节水液代谢平衡,从而发挥润肠通便作用。通过构建洛哌丁胺所致阴虚便秘大鼠模型,来考察盐炙前后知母对大鼠饮水摄食量、粪便含水率、小肠推进率、肠道通透性的变化,通过水通道蛋白(Aquaporins,AQPs)检测及结肠病理组织切片观察对润肠通便作用机理进行初步探讨。与空白组比较,模型组大鼠饮水量与摄食量在第14天均显著减少(P<0.05),大鼠粪便含水率与小肠推进率均显著降低(P<0.01),大鼠出现阴虚便秘症状,提示洛哌丁胺诱导的阴虚便秘模型成功。与模型组相比,盐知母高剂量组能够显著升高阴虚便秘大鼠饮水量摄食量(P<0.05),生知母高剂量组显著提高了阴虚便秘大鼠小肠推进率(P<0.01),盐知母高剂量组可显著提高阴虚便秘大鼠粪便含水率和小肠推进率(P<0.05);此外,盐知母高剂量组能显著提高蛋白酶的活力(P<0.05),对结肠部位纤维素活力水平亦显著提高(P<0.05);通过水通道蛋白检测发现,盐知母高剂量组能显著提高大肠AQP3和小肠AQP8水平(P<0.01),盐知母低剂量组、生知母高剂量组也可显著改善大肠AQP3的水平(P<0.05),通过病理切片观察,模型组大鼠结肠组织有明显炎性细胞浸润现象,细胞排列不规整,盐知母高剂量组、盐知母低剂量组,结肠组织损伤程度明显减少,结构基本恢复正常。知母能够提高大肠与小肠内水通道蛋白水平以及结肠部位蛋白酶的活力,进而调节阴虚便秘大鼠水液代谢平衡,改善因阴虚便秘所导致的结肠组织的炎症反应,提高阴虚便秘大鼠小肠推进率和粪便含水率,从而缓解阴虚便秘症状。在相同剂量下,盐知母与生知母相比更具有优势。To study the effect of Anemarrhena asphodeloides on Yin deficiency constipation rats before and after salt processing,and to ex-plore whether its processing can play a role in moistening intestine and purgative by regulating the balance of water metabolism.A rat model of Yin deficiency constipation induced by loperamide was established to observe the changes of water and food intake,fecal water content,small intestinal transit rate,and intestinal permeability of Anemarrhena asphodeloides before and after salt scorching in the rats.Aquaporins(AQPs)detection and pathological observation of colon tissue were used to explore the mechanism of moist intestine and defecation.On day 14,the model group's food and drink intake were significantly lower than those of the control group(P<0.05).The fecal water content and small intestine propulsion rate of the model group were significantly decreased(P<0.01),and the rats showed the symptoms of Yin de-ficiency constipation,suggesting that the model of Yin deficiency constipation induced by loperamide was successful.Compared with the model group,the high dose of Anemarrhena asphodeloides and the high dose of salt Anemarrhena asphodeloides could significantly increase the water and food intake of the rats with Yin deficiency constipation(P<0.01),and the high dose of Anemarrhena asphodeloides could significantly increase the small intestine propulsion rate of the rats with Yin deficiency constipation(P<0.01).The high dose of salt Anem-arrhena asphodeloides could significantly increase the fecal water content and small intestine propulsion rate of Yin deficiency constipation rats(P<0.05).In addition,the activity of protease and cellulose in colon were significantly increased in the high dose salt Anemarrhena as-phodeloides group(P<0.05).Aquaporins detection showed that the high dose group of salt Anemarrhena asphodeloides could significantly increase the levels of AQP3 in the large intestine and AQP8 in the small intestine(P<0.01),and the large intestine's AQP3 level could be consi
