摘 要:本研究旨在构建黄秋葵嫩果指纹图谱,建立同时测定4种成分含量的一测多评法。试验采用高效液相色谱法,建立黄秋葵嫩果主成分指纹图谱检测方法,以异槲皮苷为内参物,建立黄酮类成分槲皮素-3-O-龙胆二糖苷、槲皮素-3-O-木糖-(1-2)-葡萄糖、槲皮素-3-O-(6''-O-丙二酰基)-葡萄糖苷的一测多评法,与外标法测定结果对比,判断方法准确性。结果表明,15批黄秋葵嫩果指纹图谱共确定并鉴定4个黄酮类主成分共有峰,分别为槲皮素-3-O-龙胆二糖苷、槲皮素-3-O-木糖-(1-2)-葡萄糖、异槲皮苷、槲皮素-3-O-(6''-O-丙二酰基)-葡萄糖苷。通过一测多评法和外标法测定的4种黄酮类成分含量无显著性差异(RSD<2%)。本研究所建立的黄秋葵嫩果指纹图谱及一测多评方法准确可靠,为黄秋葵质量控制及开发利用奠定了基础。To construct the fingerprint of okra pods and establish a quantitative analysis of multi-components by single marker(QAMS)method for simultaneous determination of the contents of 4 components.High performance liquid chromatography was used to establish the fingerprint detection method of the main components of okra pods.Selecting isoquercitrin as the internal reference substance,a QAMS meth-od for the flavonoid components,Quercetin-3-O-gentiobioside,Quercetin-3-O--D-xylosyl-(1-2)--D-glucoside,and Quercetin-3-O-(6''-O-malonyl)--D-glucoside was established.And the results were compared with those obtained by the external standard method(ESM)to de-termine the accuracy of the QAMS method.The results showed that 4 common peaks of flavonoid components were identified in the fin-gerprint of 15 batches of okra pods,named Quercetin-3-O-gentiobioside,Quercetin-3-O--D-xylosyl-(1-2)--D-glucoside,isoquercitrin and Quercetin-3-O-(6''-O-malonyl)--D-glucoside.There were no significant differences between the results of QAMS and ESM for 4 flavonoid components,with all the RSD less than 2%.The established fingerprint and QAMS method are accurate and reliable,laying a foundation for quality control,development and utilization of okra.
