摘 要:本文旨在研究小陷胸汤在大鼠体内的成分及分布特征。试验采用超高效液相色谱四极杆/静电场轨道阱高分辨质谱法(UHPLC-Q Exactive Focus MS/MS)分析大鼠血浆、组织及排泄物中的化学成分,通过比对小陷胸汤、空白血浆、含药血浆、组织及排泄物的图谱差异,解析小陷胸汤经大鼠灌胃后不同时间点血浆、组织及排泄物中的化学成分及代谢产物。给药后30、60、90、120min的大鼠血浆样品中分别检出12、14、12、7种小陷胸汤的入血化学成分和3、8、3和5个入血代谢产物,其中成分包括生物碱类15种、核苷类2种、氨基酸类1种、有机酸类2种、糖醛类1种、挥发油及油脂类1种。代谢产物主要为小檗碱、药根碱、缬氨酸、烟酸等化合物,通过葡萄糖醛酸化、硫酸酯化、甲基化等代谢途径产生的代谢物,以Ⅱ相代谢为主。从大鼠的心、肝、脾、肺、肾、脑、粪和尿中分别检测到10、10、6、11、12、8、23、23种小陷胸汤的化学成分。本研究系统地分析了小陷胸汤在体内的分布特征,为其药效物质基础及作用机制研究提供了重要依据。To analyze the distribution characteristics of Xiaoxianxiong decoction in rats.UHPLC-Q Exactive Focus MS/MS was used to analyze the chemical composition in rat plasma,tissues and excreta.By comparing the differences in the chromatograms of Xiaoxianxiong decoction,blankplasma,drug-containing plasma,tissues and excreta,the chemical composition and metabolites in the plasma,tissues and excreta of Xiaoxianxiong decoction at different time points after intragastric administration in rats were analyzed.Totally 12,14,12 and 7 bloodc-hemical constituents and 3,8,3 and 5 metabolites of Xiaoxianxiong decoction were detected in the plasma samples of rats at 30,60,90 and 120 min after the administration,respectively.The chemical compositions including 15 alkaloids,2 nucleosides,1 amino acid,2 organic acid,1 glycoalkaloid,and 1 volatile oil and grease.It is mainly from the metabolites of compounds containing berberine,Jateorhizine,valine,nicotinic acidetc and their metabolic pathways were mainly glucuronidation,sulfation,methylation and others,which by phase II metab-olism.Totally 10,10,6,11,12,8,23 and 23 chemical compositions were detected in the heart,liver,spleen,lung,kidney,brain,excrement and urine,respectively.This study systematically analyzed the distribution characteristics of Xiaoxianxiong decoction in vivo,which pro-vided an important basis for the study of its pharmacodynamic material basis and mechanism of action.
