摘 要:采用低共熔溶剂(Deep eutectic solvents,DES)提取葛根中的黄酮类成分,并研究葛根黄酮对2型糖尿病db/db小鼠的降血糖作用。采用水浴加热法制备DES,超声辅助提取葛根中的黄酮类成分;以葛根素、大豆素、染料木素的含量为指标,通过单因素考察筛选最佳提取工艺;通过测定空腹血糖值(Fasting blood glucose,FBG)和口服葡萄糖耐量试验(Oral glucose tolerance test,OGTT),考察葛根黄酮调节血糖的作用。以氯化胆碱苹果酸制备的DES对葛根中黄酮类成分的提取效率最高,优化后葛根黄酮的提取工艺为:氯化胆碱苹果酸(1:3)、DES的含水量15%、料液比为30 mg/mL、提取时间25 min;葛根黄酮可以有效改善db/db小鼠的高血糖水平,并增强了葡萄糖耐受性。氯化胆碱苹果酸制备的DES可用于葛根中黄酮类成分的高效、快速提取,其DES提取液可用于改善db/db小鼠血糖水平。To extract flavonoids from Pueraria lobata using deep eutectic solvent(Deep eutectic solvents,DES)and to investigate the hy-poglycemic effect of Pueraria flavonoids on db/db mice with type 2 diabetes.DES was prepared using a water bath heating method,and fla-vonoids were extracted from Pueraria lobata with ultrasonic-assisted extraction.The optimal extraction process was screened through single-factor tests based on the content of puerarin,daidzein,and genistein.The hypoglycemic effects of Pueraria flavonoids were assessed by measuring fasting blood glucose(Fasting blood glucose,FBG)levels and conducting an oral glucose tolerance test(Oral glucose toler-ance test,OGTT).The DES composed of choline chloride and malic acid demonstrated the highest extraction efficiency for flavonoids from Pueraria lobata.The optimized extraction conditions were as follows:choline chloride-malic acid(1:3),15%water content in DES,a ma-terial-to-solvent ratio of 30 mg/mL,and an extraction time of 25 minutes.Pueraria flavonoids effectively improved hyperglycemia in db/db mice and enhanced glucose tolerance.The DES prepared from choline chloride and malic acid is suitable for the efficient and green extrac-tion of flavonoids from Pueraria lobata,and the DES extract can improve blood glucose levels in db/db mice with type 2 diabetes.
