摘 要:本研究建立元和正胃片高效液相色谱(High performance liquid chromatography,HPLC)指纹图谱,为元和正胃片质量标准提升提供依据。本试验采用C18色谱柱,乙腈-0.1%磷酸溶液进行梯度洗脱,流速:1.0 mL/min,柱温:30℃,进样量:10μ L,检测波长:254 nm。使用《中药色谱指纹图谱相似度评价系统》(2012版)进行相似度评价。利用化学模式识别方法判断元和正胃片多批次间的质量差异。建立了12批元和正胃片的HPLC指纹图谱,其相似度均大于0.98,为整体评价元和正胃片提供参考依据。共标定出24个共有峰,指认出芦荟大黄素、大黄酚、大黄酸、大黄素、龙胆苦苷、延胡索乙素、甘草苷、甘草酸铵8个成分。通过化学模式识别将12批样品分为2类,筛选出6个导致分类差异的主成分。分析发现6个主成分反应的色谱峰信息主要归属于大黄、甘草、龙胆、延胡索。建立了元和正胃片指纹图谱,可以比较全面、科学地对元和正胃片进行质量评价,为元和正胃片质量控制提供参考。A high performance liquid chromatography(High performance liquid chromatography,HPLC)fingerprint detection method for Yuanhe Zhengwei tablets was established to provide relevant basis for the quality standard of Yuanhe Zhengwei tablets.The experiment was performed on a C18 column with the gradient elution of acetonitrile-0.1%phosphoric acid solution at the flow rate of 1.0 mL/min,the col-umn temperature of 30°C,the injection volume of 10 μL and the detection wavelength of 254 nm.Evaluation of similarity using the“Evalu-ation system of similarity of chromatographic fingerprints of traditional Chinese medicine(2012 version)”.Chemical pattern recognition were performed to evaluate the quality of different batches of Yuanhe Zhengwei tablets.The HPLC fingerprints of 12 batches of Yuanhe Zhengwei Tablets were established,and their similarities were all greater than 0.98,which can provide a reference basis for the overall evalu-ation of Yuanhe Zhengwei tablets.A total of 24 common peaks were marked.Eight components including aloeemodin,chrysophanol,rhein,emodin,gentiopicroside,tetrahydropalmatine hydrochloride,liquiritin,andglycyrrhizic acid ammonium salt were identified.The 12 batches of samples were divided into 2 classes by chemical pattern recognition,and 6 principal components leading to differences in classification were screened.The analysis revealed that the information of the chromatographic peaks responded by the six principal components was ma-inly attributed to Rhubarb,Licorice,Gentiana scabra Bunge,and Corydalis Rhizoma.The fingerprint method of Yuanhe Zhengwei tablets was established,which can be used to evaluate the quality of Yuanhe Zhengwei tablets in a more comprehensive and scientific way,and pro-vide a reference for the quality control of Yuanhe Zhengwei tablets.
