摘 要:目的:观察加味温胆汤对阿霉素诱导心肌细胞自噬损伤的保护机制。方法:将健康SD雄性大鼠分为空白血清组和加味温胆汤血清组,分别予加味温胆汤8.64 g/kg及等体积蒸馏水灌胃,制备含药血清和空白血清。以阿霉素处理H9c2心肌细胞建立自噬模型,将细胞分为空白组、模型组、加味温胆汤组和3-MA(自噬抑制剂)组。除空白组外,其余组均用阿霉素诱导自噬并给予对应血清或3-MA。采用CCK-8法检测细胞增殖和活性,RT-qPCR检测LC3、P62、ATG5的mRNA表达,免疫组化检测P62和LC3B的阳性表达。结果:与空白组比较,模型组的细胞增殖和活性显著降低(P﹤0.01),LC3和ATG5 mRNA表达显著升高(P﹤0.01),P62 mRNA表达显著下降(P﹤0.01);与模型组比较,加味温胆汤组和3-MA组细胞增殖和活性显著上升(P﹤0.01),自噬水平降低。结论:加味温胆汤通过调控LC3、ATG5、P62等自噬因子的表达,显著增强细胞增殖和活性,保护心肌细胞免受阿霉素诱导的自噬损伤。Objective:To approach the protective mechanism of modified Wendan decoction on adriamycin-induced autophagic injury of cardiomyocytes.Methods:Healthy SD male rats were divided into blank serum group and modified Wendan decoction serum group,then given 8.64 g/kg modified Wendan decoction and equal volume of distilled water by gastric gavage respectively,in order to prepare medicated serum and blank serum.Autophagy model was established by treating H9c2 cardiomyocytes with adriamycin.The treated cardiomyocytes were divided into blank group,model group,modified Wendan decoction group and 3-MA(autophagy inhibitor)group.Except blank group,other groups all were induced autophagy by adriamycin,and given corresponding serum or 3-MA.The proliferation and viability of cells were detected by CCK-8 method.mRNA expressions of LC3,P62 and ATG5 were detected by RT-QCPR.The positive expressions of P62 and LC3B were detected by immunohistochemistry.Results:Compared with blank group:The proliferation and viability of cells in model group were significantly decreased(P﹤0.01);mRNA expressions of LC3 and ATG5 were significantly increased(P﹤0.01);and mRNA expression of P62 was significantly decreased(P﹤0.01).Compared with model group:The proliferation and viability of cells in modifiedWendan decoction group and 3-MA group were significantly increased(P﹤0.01),the autophagy level was decreased.Conclusion:Modified Wendan decoction can significantly enhance proliferation and viability of cells by regulating the expression of autophagy factors such as LC3,ATG5 and P62,etc.,which can protect cardiomyocytes from adriamycin-induced autophagic injury.
