摘  要:目的采用高效液相色谱法(High performance liquid chromatography,HPLC)建立藤黄檀的指纹图谱,研究其抗氧化活性的谱效关系。方法建立10批不同产地藤黄檀的HPLC指纹图谱,采用1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl radical,DPPH)法和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS]法对藤黄檀进行抗氧化活性作用,采用灰色关联度法(Grey relational analysis,GRA)和偏最小二乘回归分析法(Partial least squares regression analysis,PLSR)研究其谱效关系,进而筛选出其抗氧化物质基础。结果建立了10批不同产地藤黄檀的HPLC指纹图谱共有模式,确定了能够表征藤黄檀特征的15个共有峰。10批不同产地批藤黄檀均具有不同程度的抗氧化活性,并呈剂量依赖性。结论成功建立10批不同产地藤黄檀的HPLC指纹图谱与其抗氧化活性的谱效关系,为深入探究藤黄檀抗氧化药效物质基础和质量标准提供了依据。Objective To establish the fingerprint of Tenghuangtan(Dalbergia hancei Benth)by high performance liquid chromatography(HPLC)and to study the spectral-effect relationship of its antioxidant activity.Method HPLC fingerprints of 10 batches of Tenghuangtan(Dalbergia hancei Benth)from different origins were established,and the antioxidant activity of Tenghuangtan(Dalbergia hancei Benth)was investigated by the 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)method and 2,2-biazobis(3-ethylbenzothiazole-6-sulfonic acid)diammonium salt(ABTS)method,and its antioxidant activity was studied by the grey relational analysis(GRA)method and partial least squares regression analysis(PLSR)method was used for its spectral-effect relationship,and then its antioxidant substance base was screened out.Results The HPLC fingerprint patterns of 10 batches of Tenghuangtan(Dalbergia hancei Benth)from different origins were established,and 15 peaks were identified to characterize Tenghuangtan(Dalbergia hancei Benth).Ten batches of Tenghuangtan(Dalbergia hancei Benth)from different origins were found to possess antioxidant activities in different degrees in a dose-dependent manner.Conclusion The HPLC fingerprints of 10 batches of Tenghuangtan(Dalbergia hancei Benth)from different places were successfully established,which provided the basis for in-depth investigation of the antioxidant efficacy of Tenghuangtan(Dalbergia hancei Benth)and its quality standard.
