摘 要:目的为探究曼地亚红豆杉炮制前后差异性化学成分。方法采用超高效液相色谱串联四极杆静电场轨道阱高分辨质谱法(UHPLC-Q-Orbitrap HRMS)对曼地亚红豆杉中化学成分进行鉴定,根据高分辨质谱精确分子量及碎片离子信息、对照品、文献、在线数据库等对化学成分进行鉴定,并以炮制前后各成分的离子峰面积比作为炮制前后的变化指数进行对比;利用SIMCA软件建立曼地亚红豆杉不同炮制品的主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)模型,获得PCA图、OPLS-DA图和变量重要性投影(VIP)值,筛选曼地亚红豆杉炮制前后的差异性成分。结果共鉴定出74个成分,曼地亚红豆杉生品74个成分、酒炙品70个成分、酒浸品70个成分、酒洗品71个成分、酒蒸品71个成分、醋浸品71个成分、醋炒品71个成分、醋蒸品71个成分、蜜炙品72个成分、姜炙品70个成分。其中,18个成分通过与对照品比对鉴定。经PCA,发现不同炮制方法对曼地亚红豆杉的化学成分具有明显的差异,OPLS-DA筛选出VIP值>1的38个化学成分作为不同炮制方法的主要差异性成分。结论方法快速准确,可用于测定曼地亚红豆杉炮制前后的化学成分差异,通过比较化学成分差异可为临床疗效研究提供重要参考。Objective In order to study the different chemical constituents of Mandiya Hongdoushan(Taxus media Rehd)before and after processing.Method The chemical components were identified by ultra-high performance liquid chromatography(UHPLC-Q-Orbitrap HRMS)in Mandiya Hongdoushan(Taxus media Rehd)with quadrupole electrostatic field orbital well high resolution mass spectrometry,and the chemical components were identified according to the precise molecular weight and fragment ion information,reference products,literature,online database,etc.The ionic peak area ratio of each component before and after processing was used as the change index.Principal component analysis(PCA)and orthogonal partial least square discriminant analysis(OPLS-DA)models of different processed Mandiya Hongdoushan(Taxus media Rehd)were established by SIMCA software.The values of PCA,OPLS-DA and variable importance in the projection(VIP)were obtained to screen the different components of Mandiya Hongdoushan(Taxus media Rehd)before and after processing.Result A total of 74 components were identified in the experiment,including 74 components of Mandiya Hongdoushan(Taxus media Rehd)raw products,70 components of wine processing products,70 components of wine soaking products,71 components of wine washing products,71 wine steaming products,71 components of vinegar processing products,71 components of vinegar soaking products,71 components of vinegar steaming products,72 components of honey-prepared products and 70 components of ginger stir-fried products.Among them,18 compounds were identified by comparison with control substances.PCA showed that the chemical composition of Mandiya Hongdoushan(Taxus media Rehd)was significantly different in different processing methods.OPLS-DA screened 38 chemical components with VIP value>1 as the main difference components of different processing methods.Conclusion The results show that the method is rapid and accurate,and can be used to determine the chemical composition difference of Mandiya Hongdoushan(Taxus media Rehd
