摘 要:目的:通过网络药理学探讨葛根芩连汤治疗溃疡性结肠炎(UC)的分子靶点及其参与治疗UC的机制。方法:采用中药系统药理数据库(TCMSP)筛选葛根、黄芩、黄连、甘草的有效成分,在UniProt数据库中确定分子靶点,在DisGeNET数据库中获得疾病基因,对比获取交集。交叉基因进行蛋白互作(PPI)构建、GO功能富集分析及KEGG通路富集分析。共同靶点导入String数据库中,通过Cytoscape软件处理后得到核心靶点相互作用图,获得互作关系排名前30的靶点,对葛根芩连汤与UC的共同靶点进行GO功能富集分析,从生物过程(BP)、细胞组分(CC)、分子功能(MF)三个层面进行富集。使用R软件进行KEGG通路富集分析。结果:STAT3、RELA、TNF、IL6、MAPK3、MAPK14、TP53、AKT1、MAPK1、STAT1等基因排名靠前,为核心靶点。BP主要涉及对脂多糖的反应、对细菌来源分子的反应、对氧化应激的反应、活性氧代谢过程等方面。CC主要与真核生物的细胞膜膜筏结构、膜微区、膜结合细胞器结构、质膜穴样内陷有关。MF主要参与细胞因子受体结合、细胞因子活性、受体配体活性、信号受体激活剂活性等。筛选出富集基因数较多的通路为AGE-RAGE signaling pathway in diabetic complications信号通路、Lipid and atherosclerosis信号通路、IL-17 signaling pathway信号通路,以及一些癌症通路。结论:葛根芩连汤可能通过上述关键靶点及相关信号通路,在抗炎、抗氧化及抑制癌基因转录等方面,对UC发挥治疗作用。Objective To explore the molecular target of Gegen Qinlian Decoction in the treatment of ulcerative colitis(UC)and its mechanism in the treatment of UC through network pharmacology.Methods The active components of Gegen(Puerariae Lobatae Radix),Huangqin(Scutellariae Radix),Huanglian(Coptidis Rhizoma),Gancao(Glycyrrhizae Radix et Rhizoma)were screened using TCMSP.The molecular targets were identified in UniProt database,and the disease genes were obtained in DisGeNET database to get the intersections by comparison.Cross-gene was analyzed by protein-protein interaction(PPI)construction,GO functional enrichment analysis and KEGG pathway enrichment analysis.The common targets were imported into the String database,and the top 30 targets in the interaction relationship were obtained through the interaction diagram of the core targets obtained after processing by Cytoscape software.GO functional enrichment analysis was carried out on the common targets of Gegen Qinlian Decoction and UC.Enrichment was carried out from three levels:biological process(BP),cell component(CC)and molecular function(MF).KEGG path enrichment analysis was performed using R software.ResultsSTAT3,RELA,TNF,IL6,MAPK3,MAPK14,TP53,AKT1,MAPK1,STAT1 and other genes ranked high and were the core targets.BP mainly involved the reaction to lipopolysaccharide,the reaction to bacteria-derived molecules,the reaction to oxidative stress,and the process of active oxygen metabolism.CC was mainly related to the raft structure of eukaryotic cell membrane,membrane microdomain,membrane binding organelle structure and plasma membrane cave-like invagination.MF was mainly involved in cytokine receptor binding,cytokine activity,receptor ligand activity,and signaling receptor activator activity,etc.The pathways with a large number of enriched genes were AGE-RAGE signaling pathway in diabetic complications,lipid and atherosclerosis signaling pathway,and IL-17 signaling pathway,and some cancer pathways.Conclu-sion Gegen Qinlian Decoction may play a therapeutic role in UC
