摘 要:目的:探索低、中、高三种不同剂量三七皂苷R1对大鼠右心室心肌肥大的干预作用及其机制。方法:本研究采取随机对照法,将40只成年清洁级SD大鼠均分对照组、假早期干预组及低、中、高剂量三七皂苷R1早期干预组,每组8只。3周后通过血液动力学观察5组大鼠的右心室心功能改变,计算右心重量指数、形态学观察右心室心肌组织变化,分子生物学检测心肌肥大关键基因TRPC6 mRNA和蛋白表达水平的差异。结果:低、中、高剂量三七皂苷R1组、假早期干预组进行对比发现,右心室心肌肥大的指标右心室平均收缩压、右心室压力最大上升速率、右心室压力最大下降速率及右心室重量指数水平均高于对照组(P<0.01)。但是低、中、高剂量三七皂苷R1组的相关指标较假早期干预组显著降低(P<0.01),且三七皂苷R1剂量越高,血液动力学指标与其呈反比趋势;HE染色观察心肌形态学变化结果:低、中、高剂量三七皂苷R1组的改善情况要显著强于MCT组;除对照组外,其余4组的右心室TRPC6mRNA及蛋白相对表达水平均比正常值高(P<0.05),但假早期干预组增高状态最为明显(P<0.05),右心室TRPC6 mRNA及蛋白相对表达水平会因三七皂苷R1剂量的增加而降低。结论:低、中、高剂量三七皂苷R1都可减轻心肌肥大,改善心功能,机制可能与降低CaN/NFAT信号通路中TRPC6的表达有关,且影响与三七皂苷R1剂量呈正相关。Objective:Effects of Panax notoginseng saponins R1 on right ventricular myocardial hypertrophy in rats at three doses were investigated.Methods:this study used random control to divide 40 adult clean grade SD rats into 5 groups,specifically low,medium and high doses,panax notoginseng saponins R1 early intervention group,control group,false early intervention group,a total of 5 groups,each group 8.After 3 weeks,the changes of right ventricular cardiac function were observed by hemodynamics in 5 groups of rats.Right cardiac weight index,morphological observation of right ventricular myocardial tissue were calculated.Molecular biology was used to detect the difference of TRPC6mRNA and protein expression level of key genes of myocardial hypertrophy.Results:The index RVSP、RV±dp/dtmax and RVMI level of right ventricular myocardial hypertrophy were higher than those of control group(P<0.01).The correlation index of low,medium and high dose Panax notoginseng saponins R1 group was significantly lower than that of pseudo-early intervention group(P<0.01),and the higher R1 dose of Panax notoginseng saponins,the hemodynamics index showed the opposite trend.HE staining showed that the improvement of low,medium and high dose Panax notoginseng saponins R1 group was significantly better than that of MC group Group T;Except in the control group,Compared with the four groups,the expression levels of right ventricular TRPC6mRNA and protein were higher than normal(P<0.05),MCT group had the most significant increase(P<0.05),The relative expression of right ventricular TRPC6 mRNA and protein decreased with the increase of R1 dose of Panax notoginseng saponins.Conclusion:Low,medium and high doses of Panax notoginseng saponins R1 can reduce cardiac hypertrophy,Improving cardiac function,The mechanism may be related to the reduction of TRPC6 expression in CaN/NFAT signaling pathways,And the effect was positively correlated with the R1 dose of Panax notoginseng saponins.
