摘　　要：目的:探究二甲双胍联合姜黄素对雄激素非依赖性前列腺癌细胞PC3生物学行为的影响。方法:依据不加任何药物、加入半数抑制浓度(IC50)二甲双胍、加入IC50姜黄素、加入IC50二甲双胍及姜黄素分别记为对照组、二甲双胍组、姜黄组、联合组,对PC3细胞增殖抑制率、凋亡率、侵袭及迁移等情况进行检测,并测定PC3细胞相关蛋白情况。结果:联合组PC3细胞增殖抑制率高于二甲双胍组、姜黄组(P<0.05)。联合组PC3细胞凋亡率、Bax/Bxl-2高于二甲双胍组、姜黄组及对照组,侵袭及迁移个数、基质金属蛋白酶(MMP)-2、MMP-9低于二甲双胍组、姜黄组及对照组(P<0.05)。结论:二甲双胍联合姜黄素有效提高Bax/Bxl-2值,抑制MMP-2、MMP-9表达,从而达到抑制雄激素非依赖性前列腺癌患者PC3细胞侵袭、迁移,促使其凋亡目的,对患者预后改善有重要意义。Objective:To investigate the effect of metformin combined with curcumin on the biological behavior of androgen independent prostate cancer cell line PC3.Method:The proliferation inhibition rate,apoptosis rate,invasion,and migration of PC3 cells were detected by adding half maximal inhibitory concentration(IC50)metformin,IC50 curcumin,IC50 metformin,and curcumin as control group,metformin group,curcumin group,and combination group,respectively.The PC3 cell related proteins were also measured.Result:The inhibition rate of PC3 cell proliferation in the combined group was higher than that in the metformin group and the ginger group(P<0.05).The apoptosis rate and Bax/Bxl-2 of PC3 cells in the combined group were higher than those in the metformin group,turmeric group,and control group.The number of invasions and migrations,matrix metalloproteinase-2(MMP)-2,and MMP-9 were lower than those in the metformin group,turmeric group,and control group(P<0.05).Conclusion:Metformin combined with curcumin effectively increases Bax/Bxl-2 values,inhibits MMP-2 and MMP-9 expression,thereby achieving the goal of inhibiting PC3 cell invasion and migration in androgen independent prostate cancer patients,promoting apoptosis,and improving patient prognosis.
