摘 要:目的探讨黄精提取物对自然衰老大鼠胸腺指数、胸腺组织氧化应激指标、衰老蛋白p53、p21表达和胸腺组织病理的影响。方法将96只18月龄大鼠随机分为黄精提取物高、中、低剂量组和老年组,每组24只;另设24只2月龄大鼠为青年组。青年组和老年组灌胃等量蒸馏水,黄精提取物高、中、低剂量组分别灌胃4、2、1 g/kg的黄精提取物,持续12周。在第4、8、12周时,每组取8只大鼠,计算胸腺指数,检测胸腺组织总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平和p53、p21蛋白的表达,观察胸腺组织病理切片。结果随周龄的增加,老年组大鼠胸腺指数降低,胸腺组织T-AOC水平降低,MDA水平升高,p53和p21蛋白表达均有上调(P<0.05);胸腺组织皮质、髓质界限不明,可见脂肪空泡和炎性细胞浸润,组织胶原沉积明显。经黄精提取物干预后,老年组大鼠胸腺指数升高,胸腺组织T-AOC、SOD水平升高,MDA水平降低,p53和p21蛋白表达均有下调(P<0.05);胸腺组织病理情况有所改善。结论黄精提取物可有效调节胸腺组织氧化应激水平,延缓自然衰老大鼠的胸腺退变。Objective To observe the effects of Rhizoma Polygonati Extract(RPE)on the thymus index,oxidative stress level,the expression of p53 and p21 protein and the pathological morphology of thymus tissues in naturally aging rats.Methods 9618-month-old rats were randomly divided into 4 groups:a high-dose group,a medium-dose group,a low-dose group and an old-aged group,with 24 rats in each group.Another 242-month-old rats were set as a young-aged group.For a duration of 12 weeks,the young-aged and old-aged groups were gavaged with the same amount of distlled water,while the high-dose,medium-dose and low-dose groups were gavaged with 4g/kg,2g/kg,and 1g/kg of RPE respectively.At the 4th,8th,and 12th weeks,8 rats taken from each group were killed under anesthesia to calculate the thymus index,detect the total antioxidant capacity(T-AOC),superoxide dismutase(SOD),and malondialdehyde(MDA)levels in thymus tissue,measure the expression of p53 and p21 proteins,and observe the pathological sections of thymus tissue.Results With the increase of age,in the old-aged group the thymus index and the T-AOC level in the thymus tissue decreased,while the MDA level increased.Both expression of P53 and P21 proteins were up-regulated(P<0.05).The boundaries between the cortex and medulla of the thymus tissue were unclear,and adipose vacuoles and inflammatory cell infiltration were seen.Tissue collagen deposition was obvious.After intervention with RPE,the thymus index,as well as the levels of T-AOC and SOD in the thymus tissue increased,while the MDA level decreased.Both expressions of p53 and p21 proteins were down-regulated(P<0.05);while the pathological conditions of thymus were improved.Conclusion RPE could delay the senescence of thymus by regulating the level of oxidative stress.
