摘  要:【目的】探明不同起源银杏Ginkgo biloba古树二代家系黄酮含量,为后续银杏育种提供依据。【方法】以来自中国21个省(区、市)的277个古银杏半同胞家系幼苗为试验材料,每个家系8株幼苗,将这些幼苗种植于南京林业大学下蜀林场(32°12′N,119°22′E),给予相同的栽培管理条件。采用分光光度计法测定277个家系的叶片总黄酮含量,根据总黄酮含量,优选出黄酮含量差距较大的家系,采用高效液相色谱法分析槲皮素、山柰酚、异鼠李素的含量。【结果】银杏幼苗的地径、苗高与起源间不具有显著相关性,叶片总黄酮含量与起源极显著相关,各起源的古银杏二代苗叶片的总黄酮含量具有显著差异。其中山东起源的幼苗叶片总黄酮含量最低,平均值为(15.91±2.07)mg/g;河南起源的幼苗叶片总黄酮含量最高,平均值为(27.07±2.88)mg/g。黄酮醇苷含量与总黄酮含量的关系呈现总黄酮含量高则黄酮醇苷含量相对较低的趋势。【结论】银杏古树二代家系幼苗叶片的黄酮含量与其起源具有显著相关性,筛选潜力大。河南起源的银杏家系为筛选出的优良种质,可进行推广栽培。【Objective】The flavonoid content of the ancient second-generation family lines of Ginkgo biloba from different seed sources was explored to provide a basis for subsequent breeding of Ginkgo biloba.【Method】Seedlings of 277 ancient ginkgo semi-sibling lineages from 21 provinces(districts and cities)in China were used as test materials,with eight seedlings from each lineage,which were planted in the Xiashu Forestry Farm of Nanjing Forestry University(32°12′N,119°22′E)and given the same cultivation and management conditions.The total flavonoid content of the leaves of 277 families was determined by spectrophotometer,and based on the total flavonoid content,the families with a large difference in flavonoid content were preferred,and the contents of quercetin,kaempferol,and isorhamnetin were analyzed by high performance liquid chromatography.【Result】The ground diameter and seedling height of ginkgo seedlings were not significantly correlated with the starting seed source,and the total flavonoid content of leaves was highly significantly correlated with the starting seed source,and the total flavonoid content of leaves of second-generation ancient ginkgo seedlings from each starting seed source was significantly different.The lowest total flavonoid content was found in the seedling leaves from the seed source in Shandong,with a mean value of(15.91±2.07)mg/g;and the highest total flavonoid content was found in the seedling leaves from the seed source in Henan,with a mean value of(27.07±2.88)mg/g.The relationship between flavonol glycoside content and total flavonoid content showed a trend of relatively low flavonol glycoside content for high total flavonoid content.【Conclusion】The flavonoid content of the leaves of the seedlings of the second generation of G.biloba family lines has a significant correlation with its seed source,and the screening potential is large.The ginkgo family lines from the seed source in Henan are the excellent germplasm screened and can be popularized for cultivation.
