番茄细菌性溃疡病病原菌检测鉴定及在植株体内分布规律研究

摘  要:对密执安棒形杆菌密执安亚种(Clavibacter michiganensis subsp.michiganensis,简称Cmm)侵染番茄引起的细菌性溃疡病进行田间调查,采用胶体金试纸条和PCR方法,对田间疑似番茄细菌性溃疡病的植株及其根、茎、叶等不同组织进行Cmm检测和鉴定。结果表明:番茄苗期至成株期均可发生溃疡病;病情随生育期推移而加重,露地生产的发病程度重于设施栽培的发病程度。分离菌株均为Cmm,随病情加重,病菌随维管束扩散至全株,主要分布在番茄根部、茎秆、叶片、叶柄和果实等组织中,试纸条检测和PCR检测结果一致。A field study was conducted on the bacterial canker disease caused by Clavibacter michiganensis subsp.michiganensis(Cmm)in tomatoes.Colloidal gold test strips and PCR methods were employed to detect and identify Cmm in tomato plants suspected of bacterial canker disease,as well as in different tissues such as roots,stems,leaves and others.The results showed that the canker disease could occur from the seedling stage to the maturity stage of tomatoes.The severity of the disease worsened with the extension of the growth period,with a higher incidence observed in field-grown tomatoes compared to those cultivated in facilities.All isolated strains were identified as Cmm,and as the disease progressed,the bacteria spread through the vascular bundles to the entire plant,primarily distributing in the roots,stems,leaves,petioles,and fruits of the tomato.The results from the test strips and molecular detection were consistent.
