广枣HPLC特征图谱的建立及四种黄酮类成分的测定

摘  要:建立广枣(Ziziphus jujuba var. spinosa(Bunge)Hu ex H.F.Chow.)的HPLC特征图谱及金丝桃苷、槲皮素、山奈酚、柚皮素4种黄酮类成分的定量分析方法,采用NanoChrom ChromCore 120 C18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸水溶液,流速为1.0 mL/min,检测波长为360 nm,柱温为40℃,进样量为10μL;对HPLC特征图谱进行聚类、PCA、PLS-DA分析,同时测定金丝桃苷、槲皮素、柚皮素和山奈酚的含量。结果表明,10批广枣的HPLC特征图谱有9个共有峰,相似度在0.982~0.999。聚类结果将10批广枣分为3类:S3、S6、S10聚为一类,S2、S4、S5、S7、S8、S9聚为一类,S1单独聚为一类。PCA和PLSDA分析结果均表明金丝桃苷、山奈酚、柚皮素可以作为不同批次质量评价的潜在物质。10批广枣金丝桃苷、槲皮素、山奈酚、柚皮素含量测定结果显示,4种成分在相应的浓度范围内线性关系良好,相关系数(R2)均大于0.999 90,加样回收率在98.32%~101.07%,相对标准偏差(RSD)在2.13%~3.01%。建立的HPLC特征图谱及金丝桃苷、槲皮素、山奈酚、柚皮素的含量测定法简单可行,可用于广枣的质量控制。An HPLC characteristic map of fructus choerospondiatis(Ziziphus jujuba var.spinosa(Bunge)Hu ex H.F.Chow.)was established,and a quantitative analysis method for four flavonoid components,namely hyperoside,quercetin,kaempferol,and naringenin,was used.The NanoChrom ChromCore 120 C18(4.6 mm×250 mm,5μm)column was used,and the mobile phase was acetonitrile 0.1%phosphoric acid aqueous solution,with a flow rate of 1.0 mL/min,a detection wavelength of 360 nm,a column temperature of 40℃,and an injection volume of 10μL;Cluster,PCA,PLS-DA analysis were performed on the HPLC characteristic map,and the contents of hyperoside,quercetin,naringenin,and kaempferol were determined simultaneously.The results showed that there were 9 common peaks in the HPLC characteristic map of 10 batches of fructus choerospondiatis,with a similarity of 0.982~0.999.The clustering results divided the 10 batches of fructus choerospondiatis into three categories:S3,S6,and S10 clustered together,S2,S4,S5,S7,S8,and S9 clustered together,and S1 clustered separately.Both PCA and PLS-DA analysis results indicated that hyperoside,kaempferol,and naringenin could be potential substances for quality evaluation of different batches.The determination results for the content of hyperoside,quercetin,kaempferol,and naringenin of 10 batches of fructus choerospondiatis showed there was a good linear relationship among the four components within the corresponding concentration range,with correlation coefficients(R2)greater than 0.99990.The recovery rate was between 98.32%and 101.07%,and the relative standard deviation(RSD)was between 2.13%and 3.01%.The established HPLC characteristic map and the content determination method of hyperoside,quercetin,kaempferol,and naringenin were simple and feasible,and could be used for quality control of fructus choerospondiatis.
