摘  要:目的从分子层面评价埃氏巨球菌(Megasphaera elsdenii)对小鼠3T3-L1细胞、STC-1细胞脂代谢调节作用。方法通过用不同浓度梯度(106、107、108 CFU/mL)M.elsdenii发酵上清液分别处理STC-1细胞和诱导的小鼠脂肪3T3-L1细胞,分别对处理的细胞进行细胞周期检测,对3T3-L1细胞进行油红O染色观察成脂能力,并对成脂相关基因进行蛋白检测;对STC-1细胞释放肠道激素相关基因进行转录组水平检测。结果M.elsdenii发酵上清液抑制STC-1细胞增殖以及3T3-L1细胞增殖过程,呈剂量依赖式显著促进小鼠脂肪细胞成脂(F=37.75,P<0.0001),108 CFU/mL浓度下显著提高脂肪细胞成脂相关基因C/EBPα、PPARγ、SREBF1的表达(F=8.13、48.07、31.06,P=0.0082、<0.0001、P<0.0001);抑制小鼠小肠内分泌细胞对食欲抑制激素[peptide YY(PYY)]、胰高血糖素样肽-1(GLP-1)的释放(PYY:F=38.72,P<0.0001;GLP-1:F=27.29,P=0.0001),并显著降低GLP-1转录组水平(F=366.90,P<0.0001)。结论M.elsdenii发酵上清液可通过促进小鼠脂肪细胞成脂,促进脂质基因表达,并且抑制小鼠肠道内分泌细胞产生食欲抑制类激素从而促进机体脂肪沉积。本研究结果可为微生物干预调控宿主脂肪沉积提供理论依据。Objective To evaluate the regulatory effects of Megasphaera elsdenii on lipid metabolism in mouse 3T3-L1 and STC-1 cells at molecular level.Methods Different concentrations(106,107,108 CFU/mL)of M.elsdenii fermentation supernatant were used to treat STC-1 cells and induced mouse adipocyte 3T3-L1 cells.Cell cycle analysis was conducted on the treated cells.Oil Red O staining was performed on the 3T3-L1 cells to observe adipogenic capability,and protein detection was carried out for genes related to adipogenesis.Transcriptional levels of genes related to intestinal hormone release in STC-1 cells were also examined.Results The fermentation supernatant of M.elsdenii inhibits the proliferation of STC-1 cells and the proliferation process of 3T3-L1 cells,significantly promoting adipogenesis in mouse adipocytes in a dose-dependent manner(F=37.75,P<0.0001).At a concentration of 108 CFU/mL,it significantly enhances the expression of adipogenesis-related genes C/EBPα,PPARγ,and SREBF1(F=8.13,48.07,31.06;P=0.0082,<0.0001,P<0.0001);it inhibits the release of appetite-suppressing hormones[peptide YY(PYY)],glucagon-like peptide-1(GLP-1)from mouse intestinal endocrine cells(PYY:F=38.72,P<0.0001;GLP-1:F=27.29,P=0.0001),and significantly reduces the transcriptomic level of GLP-1(F=366.90,P<0.0001).Conclusion The fermentation supernatant of M.elsdenii can promote adipogenesis in mouse adipocytes,enhance lipid gene expression,and inhibit the production of appetite-suppressing hormones by mouse intestinal endocrine cells,thus facilitating fat deposition in the body.These findings provide a theoretical basis for microbial intervention in the regulation of host fat deposition.
