摘 要:目的探究葛根芩连汤对肠道湿热证泄泻小鼠肠道菌群及短链脂肪酸(SCFAs)的影响,揭示该方剂对肠道湿热证泄泻的疗效机制。方法采用饮食失常联合高温高湿环境建立肠道湿热证泄泻小鼠模型并分为自愈组(n=5)和治疗组(n=5),同时选取5只健康小鼠设为正常组。治疗组小鼠给予葛根芩连汤灌胃治疗。运用16S rRNA基因高通量测序技术分析小鼠肠道菌群特征,应用气相色谱—质谱联用(GC-MS)检测肠道中SCFAs含量。结果Alpha多样性分析表明,治疗组小鼠肠道菌群Shannon指数和Simpson指数均低于自愈组并接近正常组水平。在门水平上,治疗组小鼠肠道中厚壁菌门、拟杆菌门、unidentified_Bacteria比例回归至正常组水平但变形菌门比例降低;属水平上,治疗组小鼠肠道中乳杆菌属、Lachnospiraceae_NK4A136_group、Candidatus_Arthromitus、Ligilactobacillus丰度接近正常组,且Ligilactobacillus的比例显著增加(F=8.780,P<0.05)。代谢组学检测结果显示,治疗组小鼠乙酸、丁酸、戊酸水平较自愈组降低且向正常组回归。结论葛根芩连汤治疗肠道湿热证泄泻的机制可能与其促进Ligilactobacillus增多和调节肠道中SCFAs水平有关。Objective To explore the impact of Gegen Qinlian Decoction on intestinal microbiota and short chain fatty acids(SCFAs)in diarrheal mice with intestinal dampness-heat syndrome,and reveal its therapeutic mechanism.Methods The diarrheal mice model with intestinal dampness-heat syndrome was established by making a living environment with high dampness and temperature,and improper diets.The model mice were divided into treatment group(n=5)and self-healing group(n=5).Five healthy mice were included in control group.The mice in the treatment group were given Gegen Qinlian Decoction.The intestinal microbiota was studied using 16S rRNA gene high-throughput sequencing,and the contents of SCFAs were detected with GC-MS.Results Alpha diversity analysis showed that the Shannon index and Simpson index in the treatment group were lower than those in the self-healing group and closed to those in the control group.At the phylum level,the relative abundance of Firmicutes,Bacteroidota and unidentified_Bacteria in the treatment group were close to that in the control group,yet that of Proteobacteria decreased in the treatment group.At the genus level,Gegen Qinlian Decoction prompted the relative abundance of Lactobacillus,Lachnospiraceae_NK4A136_group,Candidatus_Arthromitus and Ligilactobacillus back to that of the control group,and that of Ligilactobacillus significantly increased(F=8.780,P<0.05).The GC-MS results showed that the levels of acetic acid,butyric acid and valeric acid in the treatment group were lower than those in the self-healing group,tending to that in the control group.Conclusion The mechanism of Gegen Qinlian Decoction in treating diarrhea with intestinal dampness-heat syndrome may be related to its proliferative effect on Ligilactobacillus and regulation on the levels of SCFAs.
