摘  要:目的观察植物乳杆菌SAL-L型谷氨酸合生元对氧化应激损伤模型小鼠的缓解作用并探究其作用机制。方法通过腹腔注射D-半乳糖(D-gal)建立氧化应激小鼠模型,随后将小鼠随机分为正常对照组、模型对照组、植物乳杆菌SAL组、L−谷氨酸组和植物乳杆菌SAL-L型谷氨酸合生元组。连续灌胃给药4周后,收集小鼠血浆和结肠组织,检测血浆丙二醛(MDA)、总超氧化物歧化酶(T-SOD)和氧化型谷胱甘肽(GSSG)水平,通过苏木精−伊红(HE)染色评估结肠组织病理变化。使用Western blotting技术分析结肠组织紧密连接蛋白Occludin和Claudin-1的表达。结果与正常对照组相比,模型组小鼠MDA(t=4.411,P=0.001)、GSSG(t=4.516,P=0.001)水平和T-SOD(t=4.086,P=0.002)活性显著升高,同时其结肠组织结构出现了明显的损伤。合生元干预后,小鼠血浆MDA(t=1.955,P=0.190)、T-SOD(t=0.316,P=0.993)和GSSG(t=7.455,P<0.001)水平有所改善,结肠组织病理损伤减轻,紧密连接蛋白表达上调。结论植物乳杆菌SAL-L型谷氨酸合生元能够显著缓解D-gal诱导的小鼠氧化应激损伤,其机制可能与增强抗氧化防御系统、改善肠道屏障功能有关。Objective To observe the alleviating effect of Lactobacillus plantarum SAL-L glutamate synbiotic on oxidative stress injury model in mice and explore its mechanism of action.Methods The oxidative stress mouse model was established through intraperitoneal injection of D-galactose(D-gal).Subsequently,the mice were randomly divided into the normal control group,model control group,Lactobacillus plantarum SAL group,L-glutamine acid group,and Lactobacillus plantarum SAL-L glutamic acid synbiotic group.After four weeks of continuous intragastric administration,the plasma and colon tissues were collected from the mice,and the levels of malondialdehyde(MDA),total superoxide dismutase(TSOD)and oxidized glutathione(GSSG)in the plasma were measured.The pathological changes in colon tissue were evaluated using hematoxylin-eosin(HE)staining.Western blotting was employed to analyze the expression of tight junction proteins Occludin and Claudin-1 in the colon tissue.Results Compared to the normal control group,the model group exhibited significantly increased levels of MDA(t=4.411,P=0.001),GSSG(t=4.516,P=0.001),and T-SOD(t=4.086,P=0.002).There was notable damage to the structure of colon tissue.Following synbiotic intervention,improvements were observed in the plasma levels of MDA(t=1.955,P=0.190),T-SOD(t=0.316,P=0.993)and GSSG(t=7.455,P<0.001).Furthermore,colon histopathology indicated a reduction in damage and an upregulation of tight junction protein expression.Conclusion Lactobacillus plantarum SAL-L glutamate synbiotic significantly alleviates D-gal-induced oxidative stress injury in mice.Its mechanism may be associated with the enhancement of antioxidant defense system and improvement of intestinal barrier function.
